Abstract

U radu je ispitan učinak etanolnog ekstrakta cvijeta trnine (Prunus spinosa L.) na vijabilnost i proliferaciju mišjih kancerogenih Hepa 1-6 i nekancerogenih AML 12 stanica jetre, nakon 24 i 72 sata, Trypan Blue metodom i protočnom citometrijom. Za tretman Hepa 1-6 stanica korištene su koncentracije ekstrakta cvijeta trnine od 10 do 200 μg mL-1, a za AML 12 stanice od 5 do 100 μg mL-1. Utvrđeno je da ekstrakt cvijeta trnine ima statistički značajan inhibicijski učinak na proliferaciju stanica nakon 24-satne inkubacije, kod Hepa 1-6 stanica pri koncentracijama većim od 100 μg mL-1, a kod AML 12 stanica pri svim ispitanim koncentracijama izuzev 10 μg mL-1. Tijekom perioda od 72 sata kod obje stanične linije pri svim ispitanim koncentracijama utvrđen je inhibitorni učinak ekstrakta cvijeta trnine. Iz izračunatih i uspoređenih IC50 vrijednosti, ekstrakt cvijeta trnine pokazao je jači inhibitorni učinak na AML 12 staničnu liniju. Protočnom citometrijom utvrđeno je da ekstrakt cvijeta trnine uzrokuje nekrozu stanica kod obje stanične linije.

Abstract (english)

In this study effects of the blackthorn flower (Prunus spinosa L.) extract on cell proliferation and viability were examined in mouse cancer Hepa 1-6 and normal AML 12 liver cell lines using Trypan Blue method and flow cytometry after 24 and 72 hours. Different concentrations of blackthorn flower extract were used in treatment of cells (for Hepa 1–6 10-200 μg mL-1, and for AML 12 5-100 μg mL-1). Statistically significant decrease of Hepa 1-6 cell proliferation was detected at concentrations greather than 100 μg mL-1 and AML 12 cell proliferation at all examined concentrations, excluding 10 μg mL-1, after 24 hours. Statistically significant decrease of Hepa 1–6 and AML 12 cell proliferation was detected after 72 hours at all concentrations. According to calculated and compared IC50 values blackthorn flower extract has shown stronger inhibitory effect on AML 12 cell line. Results obtained with flow cytometry have shown that blackthorn flower extract causes necrosis of both cell lines.
