Abstract

U ovom radu ispitan je učinak neplanarnog PCB-153 (10-100 μM) na proliferaciju CHO (Chinese Hamster Ovary)-K1 stanica metodama Neutral Red, Kenacid Blue, MTT i Trypan Blue nakon 6 i 48 h tretmana. Metodom protočne citometrije određen je postotak živih i mrtvih stanica, kao i postotak stanica u apoptozi. Nakon 6 h tretmana značajan inhibicijski učinak CHO-K1 stanica određen je kod tri provedene metode pri koncentracijama PCB-153 od 75 i 100 μM, izuzev kod metode Kenacid Blue. Nakon 48 h PCB-153 je značajno inhibirao staničnu proliferaciju pri najvišim koncentracijama (75 i 100 μM) kod sve četiri provedene metode. Metodom protočne citometrije utvrđeno je da se proporcionalno koncentraciji PCB-153 povećava broj stanica u apoptozi nakon 6 h tretmana (12,18 % - 28,15 %), a nakon 48 h broj mrtvih stanica (5,75 % - 33,43 %).

Abstract (english)

The aim of this study was to examine the cytotoxic effects of the non-planar PCB-153 (10-100 μM) by monitoring CHO (Chinese Hamster Ovary)-K1 cell proliferation using Neutral Red, Kenacid Blue, MTT and Trypan Blue methods after 6 and 48 hours of treatment. Flow cytometry was used to distinguish live, dead and apoptotic cells. Statistically significant inhibition was determined with 75 and 100 μM PCB after 6 hours with three methods used, except Kenacid Blue. Also, statistically significant decrease of cell proliferation was detected with the higest PCB-153 concentrations (75 and 100 μM) after 48 hours with all four applied methods. The flow cytometry analysis revealed that increase in PCB-153 concentration proportionaly increased the number of apoptotic cells after 6 hours (12,18 % - 28,15 %) and number of dead cells (5,75 % - 33,43 %) after 48 hours of treatment.
