Abstract

Proizvodnja bioetanola na škrobnim sirovinama nije ekonomična te se javila potreba za novim, obnovljivim sirovinama poput lignoceluloznih materijala. Lignocelulozne sirovine složenog su sastava, a građene su od celuloze, hemiceluloze i lignina, spojeva koje kvasac ne može metabolizirati radi čega je prvo nužno provesti predtretman, to jest hidrolizirati sirovine. Hidrolizom ne dolazi samo do oslobađanja fermentabilnih šećera nego i do nastanka slabih kiselina i derivata furana koji djeluju kao inhibitori rasta. Zbog toga je za proizvodnju bioetanola na lignoceluloznim hidrolizatima potreban proizvodni soj kvasca koji daje visoki prinos etanola, ali koji je također i otporan na inhibitore rasta. U ovome radu prirodni izolat kvasca, koji proizvodi bioetanol na lignoceluloznim hidrolizima, transformiran je plazmidima koji omogućuju pojačanu ekspresiju gena za koje utvrđeno da povećavaju rezistenciju na inhibitore rasta, a prisutnost plazmida potvrđena je molekularnom analizom hibridizacijom po Southern-u.

Abstract (english)

The production of bioethanol on raw starch materials is not economical and there is a need for renewable raw materials such as lignocellulose. Lignocellulose is composed of complex compounds such as cellulose, hemicellulose and lignin. These compounds can not be directly metabolized by yeast. Therefore, it is first necessary to hydrolyze raw materials during pretreatment. Unfortunately, hydrolysis does not release only fermentable sugars, but it also releases weak acids and furan derivatives which act as growth inhibitors. Because of this, production of bioethanol on lignocellulosic hydrolysates requires adapted yeast strain(s). Those strains give high yields of ethanol and they are also resistant to growth inhibitors. In this thesis, a natural yeast isolate which produces bioethanol on lignocellulosic hydrolates, is transformed with plasmids, that allow overexpression of genes shown to increase resistance to growth inhibitors, and the presence of plasmids is confirmed by Southern blotting.
