Abstract

Da bi probiotički pripravci pokazali pozitivne učinke na zdravlje potrošača, moraju sadržavati visoku koncentraciju živih stanica. Zbog toga se istražuju različiti matriksi i metode kako bi se povećalo njihovo preživljavanje tijekom proizvodnje, čuvanja i primjene u različitim prehrambenim proizvodima te nakon oralne primjene u gastrointestinalnom traktu domaćina. Cilj ovog rada bio je ispitati utjecaj mikroinkapsulacije na preživljavanje bakterija Lactobacillus plantarum D13 i Lactococcus... More lactis ZG7-10. Stoga je provedena mikroinkapsulacija primjenom tri nosača, alginata, κ-karagenana i kazeina uz djelovanje transglutaminaze, a zatim i liofilizacija mikroinkapsuliranih stanica uz upotrebu različitih lioprotektora. Mikroinkapsulacija u alginatu i dodatak obranog mlijeka kao lioprotektora, pokazali su se najučinkovitiji za oba soja. Prolaskom mikroinkapsuliranih liofiliziranih stanica kroz simulirane uvjete gastrointestinalnog trakta, visok postotak preživjelih stanica ukazuje na zaštitni učinak mikroinkapsulacije za probiotičke sojeve, pri čemu je broj živih stanica bio veći od 108CFU/g, što je u skladu sa zahtjevima koje moraju ispunjavati probiotički proizvodi. Abstract (english)

In order to express their benefits on human health, the probiotic preparations need to contain high concentration of viable cells. Therefore, different matrices and methods have been investigated to improve their survival during production, storage or application in different food products, and after an oral application in the host gastrointestinal conditions. The aim of this thesis was to investigate influence of microencapsulation on the survival of probiotic bacteria Lactobacillus... More plantarum D13 and Lactococcus lactis ZG7-10. Therefore, the microencapsulation was performed with application of three different carriers, alginate, κ –carrageenan and transglutaminase-induced casein, and then lyophilization of microencapsulated cells with the addition of various lyoprotectors. Microencapsulation in alginate, and addition of skim milk as lyoprotector, proved to be most effective for both bacterial strains. After transit through simulated gastrointestinal tract conditions, high percentage of viable cells indicates the protective effect of the microencapsulation on the probiotic strains, wherein the number of viable cells was above 108 CFU/g, what is in accordance with the claims set up for probiotic products.
