Abstract

U ovom radu ispitan je učinak različitih koncentracija resveratrola (2,5-20 μM) pri citotoksičnom djelovanju 75 μM PCB-a 77 ili 75 μM PCB-a 153 na CHO (Chinese Hamster Ovary) – K1 stanice metodama Neutral Red, Kenacid Blue te metodom protočne citometrije. Utvrđeno je statistički značajno smanjenje vijabilnosti stanica u odnosu na kontrolni uzorak pri tretmanu CHO-K1 stanica s PCB-em 77 i PCB-em 153 metodama Neutral Red i Kenacid Blue. Blagi protektivni učinak resveratrola (u koncentracijama 2,5, 5 i 10 μM) je primijećen pri citotoksičnom djelovanju PCB-a 153, no nije se pokazao statistički značajnim. Toksični učinci dioksinu sličnog kongenera-PCB-a 77 na stanice ovarija nisu umanjeni, čak suprotno - djelovanjem resveratrola izraženiji je učinak na pad vijabilnosti stanica. Sve primijenjene in vitro metode pokazale su korespondirajuće rezultate.

Abstract (english)

In this study, the effects of different resveratrol concentrations (2.5-20 μM) on cytotoxic activity of 75 μM PCB 77 or 75 μM PCB 153 on CHO (Chinese Hamster Ovary) - K1 cells were determined by Neutral Red method, Kenacid Blue method and by flow cytometry. Statistically significant reduction in cell viability was observed after treatment of CHO-K1 cells with PCB 77 or PCB 153 in comparison to the control sample determined with Neutral Red and Kenacid Blue methods. The slight protective effect of resveratrol (at concentrations of 2.5, 5 and 10 μM) on cytotoxic activity induced by PCB 153 was observed, but results were not statistically significant. Toxic effects of dioxin-like congener-PCB 77 on ovarian cells were not reduced by the action of resveratrol, on the contrary - resveratrol has shown a more pronounced effect on the reduction in cell viability. All applied in vitro methods have shown to be consistent within results.
