In vitro istraživanje adjuvantnog učinka quisinostata na citotoksičnost 5-fluorouracila na tumorske stanice mliječne žlijezde miša

Abstract

Tumori su osnovni predmet istraživanja velike većine eksperimentalnih radova zbog pojave rastuće smrtnosti, neovisno o dobi i spolu. Tumor dojke najzastupljeniji je oblik kod ženske populacije te je kao takav predmet istraživanja ovog rada, s ciljem ispitivanja osjetljivosti stanica tumora mliječne žlijezde miša na primijenjeni lijek 5-fluorouracil (5-FU) te pomoćno sredstvo quisinostat (inhibitor histon deacetilaza), u obliku 2D i 3D staničnih kultura. Pritom, odreĎena je koncentracija lijeka koja daje približno 50% preživljenja stanica te efikasnost istovremenog tretmana s 5-FU i qusinostatom te predtretmana qusinostatom uz dodatak lijeka. Ispitan je utjecaj različitih koncentracija lijeka 5-FU te koncentracije qusinostata. Za odreĎivanje 2D staničnih kultura korišten je resazurin test, dok su za 3D stanične kulture korišteni CellTox™ Green Cytotoxicity, RealTime-Glo MT Cell Viability i CellTiter-Glo 3D Cell Viability test. Zaključno, 3D stanična kultura pokazala je veću osjetljivost na istovremeni tretman u odnosu na 2D staničnu kulturu, koji se pokazao kao najbolja opcija primjene u tretiranju stanica tumora mliječne žlijezde miša.

Abstract (english)

Tumors are the main subject of research in the vast majority of experimental studies due to the onset of increasing mortality, regardless of age and gender. Breast cancer is the most prevalent form in the female population, and as such is the subject of research in this study, with the aim of examining the sensitivity of mouse mammary gland tumor cells to the drug 5-fluorouracile (5-FU) and the adjuvant quisinostat (histone deacetylase inhibitor), in the form of 2D and 3D cell cultures. The concentration of the drug giving approximately 50% cell survival and the efficacy of combination treatment with 5-FU and quisinostat and pretreatment with qusinostat with drug addition were determined. The effect of different concentrations of 5-FU and the concentrations of qusinostat were examined. Resazurin assay was used to determine viability in 2D cell cultures, whereas CellTox ™ Green Cytotoxicity, RealTime-Glo MT Cell Viability and CellTiter-Glo 3D Cell Viability Assay were used for 3D cell cultures. To conclude, 3D cell culture showed greater sensitivity to combination treatment than 2D cell culture, which has proven to be the best application option in the treatment of mouse mammary gland tumor cells.
