Abstract

Karcinom štitnjače najčešći je maligni endokrini tumor. Mnogo gena je uključeno u njegov nastanak, a jedan od njih je protoonkogen RET koji kodira za transmembranski receptor RET. Točkaste mutacije protoonkogena RET te njegove fuzije s različitim genima uzrokuju nastajanje mutiranog proteina RET. Kada se na mutirani protein RET vežu faktori rasta, pokreću se različiti signalni putevi kao što su MAPK i PI3K/Akt/mTOR signalni put, čija stalna aktivnost dovodi do nekontroliranog rasta stanica i inhibicije apoptoze te na taj način nastaje tumor. Ovi tumoria se dijagnosticiraju opipom i ultrazvukom, a njihovo liječenje se provodi operacijom, terapijom radioaktivnim jodom ili inhibitorima tirozin kinaza (TKI).

Abstract (english)

Thyroid carcinoma is the most common endocrine malignancy. Many genes are involved in its formation and one of them is RET protooncogene, which encodes a transmembrane receptor RET. Mutant protein RET is created by point mutations or RET fusions. Protein RET binding growth factors will cause abnormal activation of signal pathways, which include MAPK and PI3K/Akt/mTOR signal pathways. Their constant activity leads to uncontrollable cell growth, inhibition of apoptosis and tumour formation. Thyroid cancer diagnosis can be done by palpation or ultrasound and treatment options are operation, radioactive iodine therapy and thyroid kinase inhibitors (TKI).
