Abstract

RNA egzosom je multiproteinski kompleks ključan za kontrolu kvalitete RNA u svim eukariotskim stanicama. Egzoribonukleazna podjedinica RNA egzosoma Rrp6 esencijalna je za mejozu i sporulaciju kvasca S. cerevisiae, međutim razina proteina Rrp6 smanjuje se tijekom mejoze. U ovom je radu pokazan proteolitički mehanizam regulacije razine proteina Rrp6 kompleksom koji promovira anafazu/ciklosom (APC/C) i proteasomom. Rezultati su pokazali kako se u uvjetima mejoze razina Rrp6 stabilizira u mutantu kvasca u kojem je onemogućeno vezanje APC/C na Rrp6 te u uvjetima rasta u kojima je proteasom inhibiran. Također, degradacija Rrp6 tijekom mejoze dovodi do stabilizacije nekodirajućih RNA transkripata (MUT) kojima su uloge slabo istražene, zbog čega je razmotrena i potencijalna regulatorna uloga antisense transkripta s lokusa RRP6, MUT1312, u regulaciji ekspresije Rrp6. Ovim radom istražen je i fiziološki značaj degradacije Rrp6 tijekom mejoze, pri čemu je pokazano da zadržavanje visoke razine proteina Rrp6 tijekom mejoze ima negativan učinak na tijek sporulacije.

Abstract (english)

The RNA exosome is a multiprotein complex crucial for RNA quality control in all eukaryotic cells. The exoribonuclease subunit of RNA exosome Rrp6 is essential for meiosis and sporulation in the yeast S. cerevisiae, however Rrp6 protein level decreases during meiosis. This work demonstrates that Rrp6 level is regulated through targeted proteolysis by the anaphase promoting complex/cyclosome (APC/C) and the proteasome. The results show that Rrp6 is stabilized in meiotic conditions when the binding site of APC/C is mutated or when the proteasome is inhibited. As degradation of Rrp6 during meiosis results in stabilization of non-coding RNA transcripts (MUTs) with poorly characterized roles, this work explored the potential role of the antisense RRP6 transcript, MUT1312, in regulation of Rrp6 expression. The physiological significance of Rrp6 degradation was also examined, showing that maintaining high Rrp6 protein levels throughout meiosis had a negative effect on sporulation kinetics.
