Characterisation of filamentous fungi isolated from noble rotten grape berries

Abstract

Botrytised wines are wines that have been produced by adding noble rotten berries during fermentation. Main filamentous fungi responsible for the noble rot of grape berries is Botrytis cinerea. However, on grape berries B. cinerea is accompanied by other indigenous filamentous fungi that could affect the noble rotting process. Those fungi need to be identified to research the relationship between them and the progression of the noble rot process. Main purpose of this thesis was to identify selected isolates of filamentous fungi using morphology and molecular methods such as ARDRA and ITS sequencing. MALDI-ToF method was tried to determine if it could be used for reliable identification and RAPD method was used to determine if there is any genetic variance between the Botrytis isolates. Identified fungi were B. cinerea, fungi from the genus Alternaria, Cladosporium, Aspergillus and Penicillium. MALDI-ToF proved to be a good alternative as a method, although it is not as effective as ITS sequencing and the RAPD method indicated that there might be some genetic variability between the B. cinerea isolates.

Abstract (croatian)

Botritizirana vina su vina koja su bila proizvedena dodavanjem bobica grožđa plemenite truleži tijekom fermentacije. Glavna filamentozna gljiva odgovorna za plemenito truljenje grožđa je Botrytis cinerea. Međutim, na bobicama grožđa prisutne su i druge filamentozne gljive zajedno sa B. cinerea i one bi mogle imati utjecaj na proces plemenitog truljenja. Potrebno je identificirati te gljive kako bi se moglo provesti istraživanje utjecaja tih gljiva i progresije procesa plemenitog truljenja. Cilj ovog diplomskog rada je bila identifikacija izolata filamenoznih gljiva koristeći morfologiju i molekularne metode poput ARDRA i ITS sekvencioniranje. MALDI-ToF metoda je isprobana kako bi se odredilo da li bi se mogla koristiti za pouzdanu identifikaciju i RAPD metoda je bila korištena kako bi se odredilo da li postoji genetička razlika između izoliranih Botrytis sojeva. Identificirane gljive su bile B. cinerea, gljive roda Alternaria, Cladosporium, Aspergillus i Penicillium. MALDI-ToF se pokazala kao dobra alternativna metoda, iako nije bila efektivna kao ITS sekvencionijanje. RAPD metoda je ukazala da može biti neka genetička raznolikost između izolata B.cinerea.
