Abstract

U području nutricionizma, sistemsko proučavanje bioloških sustava omogućuje bolje razumijevanje utjecaja prehrane na zdravlje. Sistemsko pristupanje prehrani omogućuje personalizirane preporuke temeljene na individualnim potrebama, genetskim čimbenicima i specifičnim metaboličkim odgovorima. U ovom radu je prikazan i analiziran matematički model metabolizma fruktoze u programskom paketu CellDesigner. Glavni cilj istraživanja bio je ispitati kako unos različitih obroka, s obzirom na količinu fruktoze, utječe na metabolizam masti povezan s razvojem nealkoholne masne bolesti jetre. Provedene su simulacije modela koristeći različite početne vrijednosti fruktoze pri različitim obrocima (doručak, marenda, ručak, užina i večera). Rezultati ukazuju da povećane koncentracije fruktoze u prehrani doprinose povećanju nakupljanja triglicerida u jetri. Provedena je analiza lokalne parametarske osjetljivosti u kojoj su se gliceraldehid-3-fosfat i piruvat pokazali kao najosjetljivije varijable modela. Model uspješno simulira promjenu koncentracije fruktoze i njenih metabolita.

Abstract (english)

In the field of nutrition, the systemic study of biological systems allows for a better understanding of the impact of nutrition on health. A systemic approach to nutrition enables personalized recommendations based on individual needs, genetic factors and specific metabolic responses. This paper presents and analyzes the mathematical model of fructose metabolism in the CellDesigner software package. The main goal of the research was to examine how the intake of different meals, with regard to the amount of fructose, affects fat metabolism associated with the development of non-alcoholic fatty liver disease. Model simulations were carried out using different initial values of fructose at different meals (breakfast, brunch, lunch, snack and dinner). The results indicate that increased concentrations of fructose in the diet contribute to increased accumulation of triglycerides in the liver. An analysis of local parametric sensitivity was performed, in which glyceraldehyde-3-phosphate and pyruvate proved to be the most sensitive variables of the model. The model successfully simulates the change in the concentration of fructose and its metabolites.
