Abstract

U ovom radu provedena je alkalna predobrada trstike (γ0 = 40 g L⁻¹; wNaOH = 1 – 6 %) pri relativno niskoj temperaturi od 60 °C tijekom 3 h, kao i alkalna predobrada trstike (wNaOH = 1 %) u visokotlačnom reaktoru pri temperaturi od 180 °C tijekom 10 minuta. Za hidrolizu predobrađene trstike korišteni su komercijalni enzimi u liofiliziranom obliku (Cellulase from Aspergillus niger, Hemicellulase from Aspergillus niger) i u tekući enzimi (Cellulase, enzyme blend, Viscozyme L). Ispitivanje učinkovitosti enzimske hidrolize provedeno je pri temperaturama od 35, 40 i 45 °C pri kojima može rasti veliki broj mikroorganizama producenata različitih biokemikalija i biogoriva. Odabir ovih temperatura za provođenje procesa hidrolize omogućuje jednostavnije uspostavljanje procesa simultane saharifikacije i fermentacije. Učinkovitost hidrolize glukana i ksiliana dostupnih u predobrađenoj trstici bila je u rasponu od 60 do 100 %, a hidroliza glukana i ksilana od 100 % postignuta je kod hidrolize s 1 mL Cellulase, enzyme blend i 1,5 mL Viscozyme L.

Abstract (english)

In this work, alkaline pretreatment of reed (γ0 = 40 g L⁻¹; wNaOH = 1 – 6 %) was carried out at a relatively low temperature of 60 °C for 3 hours, as well as alkaline pre-treatment of reed (wNaOH = 1%) in a high pressure reactor at a temperature of 180 °C for 10 minutes. Commercial enzymes in lyophilized form (Cellulase from Aspergillus niger, Hemicellulase from Aspergillus niger) and liquid enzymes (Cellulase, enzyme blend, Viscozyme L) were used for the hydrolysis of pre-treated reed. The effectiveness of enzymatic hydrolysis was tested at temperatures of 35, 40, and 45 °C, at which a large number of microorganisms producing various biochemicals and biofuels can grow. The selection of these temperatures for carrying out the hydrolysis process facilitates the implementation of simultaneous saccharification and fermentation processes. The efficiency of hydrolysis of glucans and xylans available in the pre-treated reed was from 60 to 100 %, with 100 % hydrolysis of glucans and xylans achieved with 1 mL of Cellulase, enzyme blend, and 1.5 mL of Viscozyme L.
