Abstract

Psoriasis is a chronic, systemic, immune-mediated disease characterised by keratinocyte hyperproliferation, infiltration of immune cells into the dermis and epidermis and an excessive inflammatory signaling that is creating a positive loop. In this study we aimed to assess the role of NLRP6 and GSDMD in the pathophysiology of psoriasis, using an imiquimod murine model of psoriasis. Bioinformatic analysis confirmed the already reported involvement of inflammasome and pyroptosome complexes in human psoriasis and mouse imiquimod induced psoriasis-like inflammation. We confirmed that there is an induced pro- and anti inflammatory response in in vitro bone marrow-derived macrophages activated with imiquimod (IMQ). In vivo study suggested there might be some differential trends between wild type and knockout mice treated with IMQ, however these trends would have to be evaluated by repeating the study with a greater number of mice. Future research should involve in vitro HaCaT culture and flow cytometry analysis.

Abstract (croatian)

Psorijaza je kronična, sistemska i imunološki posredovana bolest karakterizirana hiperproliferacijom keratinocita, infiltracijom imunoloških stanica u dermis i epidermis te pretjeranim upalnim signaliziranjem koje stvara pozitivnu spregu. U ovom smo istraživanju željeli odrediti ulogu gena NLRP6 i GSDMD u patofiziologiji psorijaze, koristeći imikvimod model psorijaze na miševima. Bioinformatička analiza potvrdila je već zabilježenu uključenost inflamasoma i piropotosoma u ljudskoj psorijazi i mišjoj upali induciranoj imikvimodom. Potvrdili smo da postoji pro- i anti-upalni odgovor u in vitro kulturi makrofaga izoliranih iz koštane srži i aktiviranih imikvimodom (IMQ). In vivo istraživanje sugeriralo je da postoje mogući različiti trendovi između divljih i knockout miševa tretiranih sa imikvimodom. Međutim, spomenuto je potrebno dodatno procijeniti ponavljanjem istraživanja s većim brojem miševa. Buduća istraživanja trebala bi uključiti in vitro HaCaT kulturu i analizu protočnom citometrijom.
