Abstract

rava iva (Teucrium montanum L.) je mediteranska biljna vrsta čija se duga povijest tradicionalne upotrebe u prevenciji i liječenju različitih oboljenja pripisuje biološki aktivnim spojevima, poput polifenolnih spojeva. Uzevši u obzir nedostatak literaturnih podataka o valorizaciji bioaktivnog sastava trave ive, cilj ovog rada bio je inkapsulirati polifenolni ekstrakt trave ive u formi nanovlakana korištenjem binarnih sustava polimernih nosača na bazi pululana i hidroliziranog kolagena, odnosno pululana i zeina, u omjerima 80:20, 60:40 i 40:60, primjenom tehnike elektroispredanja. Formuliranim nanovlaknima određena je učinkovitost inkapsulacije ukupnih polifenola spektrofotometrijskom metodom te pojedinačnih feniletanoidnih glikozida tekućinskom kromatografijom visoke djelotvornosti (HPLC-PDA), kao i morfološke karakteristike. Ehinakozid, teupoliozid, stahizozid A, poliumozid i verbaskozid određeni su kao najzastupljeniji polifenolni spojevi u ekstraktu trave ive. Osim formulacije PUL:KOL/40:60, sve preostale kombinacije nosača rezultirale su uspješno proizvedenim nanovlaknima ekstrakta trave ive, s inkapsulacijskom učinkovitošću iznad 80 %. Kombinacije pululana i zeina rezultirale su većim prosječnim promje- rom nanovlakana (131,60 – 187,16 nm) u odnosu na binarne sustave pululana i hidroliziranog kolagena (115,89 – 120,57 nm).

Abstract (english)

Mountain germander (Teucrium montanum L.) is a Mediterranean plant whose long traditional use for the prevention and treatment of various diseases is attributed to biologically active compounds, such as polyphenolic compounds. Considering the lack of literature data on the valorization of bioactive composition of mountain germander (MG), the aim of this work was to encapsulate the MG polyphenolic extract in the form of nanofibers using binary polymer carrier systems based on pullulan and hydrolyzed collagen, i.e. pullulan and zein, in ratios of 80:20, 60 :40 and 40:60, using electrospinning technique. The encapsulation efficiency of the total polyphenols by spectrophotometric method and the individual phenylethanoid glycosides by high performance liquid chromatography (HPLC-PDA) as well as the morphological properties were determined using the formulated nanofibers. Echinacoside, teupolioside, stachysoside A, polyumoside and verbascoside were determined to be the most abundant polyphenolic compounds in the MG extract. With the exception of the PUL:KOL/40:60 formulation, all other carrier combinations resulted in successfully produced nanofibers from MG extract, with an encapsulation efficiency of over 80 %. Combinations of pullulan and zein resulted in a larger average diameter of nanofibers (131.60 – 187.16 nm) compared to the binary system of pullulan and hydrolyzed collagen (115.89 – 120.57 nm).
