Abstract

Punila su praškaste tvari koje se koriste u farmaceutskoj industriji za povećanje mase tableta i njihove uniformnosti. Ona moraju biti fiziološki inertna, kompatibilna s aktivnom tvari i kemijski stabilna. Cilj ovog rada bio je pratiti fizikalna svojstva dva punila (škrob i laktoza) pri različitim udjelima vlage. Oba praha kondicionirana su na različite sadržaje vlage, u rasponu 0-15 % za laktozu i 0-30 % za škrob. Stvarni udio vlage određen je gravimetrijski. Mjerena fizikalna svojstva su aktivitet vode, boja, Feretov promjer, površina čestica te slobodna i vibraciona nasipna gustoća. Uzorcima su snimani NIR spektri te je primjenom metode parcijalnih najmanjih kvadrata (engl. Partial Least Squares, PLS) utvrđivana je povezanost udjela vlage, promatranih fizikalnih svojstava i NIR spektara. Povećanje udjela vlage drugačije utječe na navedena fizikalna svojstva škroba i laktoze što se odražava na NIR spektre. Promjene u NIR spektrima uspješno su analizirane primjenom PLS metode.

Abstract (english)

Diluent excipients are powdery substances which are used as pharmaceutical ingredients to increase the weight and uniformity of tablets. Diluents must be physiologically inert, compatible with the active pharmaceutical ingredient and chemically stable. The aim of this work was to examine physical properties of two specific diluents (starch and lactose) at different moisture contents Both powders were conditioned to different moisture contents in the ranges 0-15% for lactose and 0-30 % for starch. Their moisture contents were determined via loss on drying. The physical properties measured were water activity, color, Feret diameter, particle area, poured and tap bulk densities. NIR spectra of all samples were recorded. The connection between moisture content, observed physical properties and NIR spectra was determined using the Parital Least Squares method (PLS). Increasing moisture affects certain physical properties more than other, both in lactose and starch, and those changes can be noticed in NIR spectra. The changes in NIR spectra were succesfully analyzed using PLS method.
