Abstract

U ovom radu ispitani su in vitro učinci 5 – 100 µM resveratrola (RSV) i kurkumina (CRC), kao i ferocenom modificiranih polifenola – derivata resveratrola (RF) i analoga kurkumina (CF) na humane keratinocite (HaCaT). CRC je pokazao najizraženiji citotoksičan učinak (metode Trypan Blue i MTT), dok je antiproliferativan učinak CF najmanje izražen (IC50 > 100 μM). Svi ispitani spojevi smanjili su oksidacijski stres induciran s tBHP (tert-butil hidroperoksid), pri čemu je antioksidacijska aktivnost najizraženija za CRC i CF. Analiza stanične smrti protočnom citometrijom pokazala je najveći udio stanica u apoptozi i nekrozi pri tretmanu keratinocita s CRC, dok je autophagy induction ratio bio najviši za stanice tretirane sa 100 µM CRC i iznosio je 1,67 (p < 0,001). Uvođenje ferocena u strukture polifenola dovelo je do izmjenjene biološke aktivnosti novosintetiziranih spojeva, pri čemu je njihova citotoksičnost značajno niža u odnosu na RSV i CRC, a antioksidacijska aktivnost zadržana.

Abstract (english)

In this study, the in vitro effects of 5-100 µM resveratrol (RSV) and curcumin (CRC) as well as ferrocene-modified polyphenols - resveratrol derivatives (RF) and curcumin analogs (CF) - on human keratinocytes (HaCaT) were examined. CRC showed the most pronounced cytotoxic effect (Trypan Blue and MTT methods), while the antiproliferative effect of CF was the least pronounced (IC50 > 100 μM). All tested compounds reduced oxidative stress induced by tBHP (tert-butyl hydroperoxide), with antioxidant activity being most pronounced for CRC and CF. Analysis of cell death by flow cytometry showed the highest percentage of cells in apoptosis and necrosis in keratinocytes treated with CRC, while the autophagy induction ratio was the highest in cells treated with 100 µM CRC and was 1.67 (p < 0.001). The introduction of ferrocene into the polyphenol structures resulted in altered biological activity of the newly synthesized compounds, with their cytotoxicity being significantly lower compared to RSV and CRC, while antioxidant activity was maintained.
