Abstract

Auksini su biljni hormoni koji su po kemijskoj strukturi indolni spojevi, a kao takvi mogu posjedovati antikancerogena svojstva. Primjena ove vrste spojeva umnogome bi olakšala terapiju raka jer je riječ o prirodnim i neinvazivnim spojevima prisutnim u biljkama, mikroorganizmima i čovjeku. U ovom radu istraženo je citotoksično, prooksidativno i mutageno djelovanje auksina (indol-3-octena kiselina, 2-metil indol-3-octena kiselina, indol-3-maslačna kiselina i indol-3-propionska kiselina) na stanice raka debelog crijeva (Caco-2). Niti jedan istraživani auksin nije pokazao citotoksični učinak. Otkriveno je da indol-3-octena kiselina (0,01 mM) najintenzivnije djeluje na povećanje stvaranja slobodnih radikala (za 274,89 %), dok peroksidaciju lipida u najvećoj mjeri uzrokuje indol-3-maslačna kiselina (0,5 mM). Niti jedan od auksina nije pokazao mutageno djelovanje. Tretman s indol-3-propionskom kiselinom pokazao je najbliži rezultat onom koji nastaje nakon tretmana s vodikovim peroksidom.

Abstract (english)

Auxins are plant hormones which are, by their structure, indolic compounds and with that comes a possibility that they can have anticancerogenic properties. Application and implementation of these compounds could ease the cancer therapy in huge extent because they are noninvasive, naturaly occuring in plants, microorganisms and humans. In this thesis cytotoxic, prooxidative and mutagenic effect of auxins (indole-3-acetic acid, 2-methyl indole-3-acetic acid, indole-3-butyric acid and indole-3-propionic acid) on human colorectal adenocarcinoma cells (Caco-2) were investigated. None of the tested auxins did show cytotoxic effect. It was discovered that IAA (0,01 mM) caused the highest formation of free radicals (for 274,89 %). On the other hand, the highest peroxidation of lipids was caused by IBA (0,5 mM). None of the auxins did show mutagenic effects. Treatment with indole-3-propionic acid showed the result closest to the one observed after the treatment with hydrogen peroxide.
