Abstract

Međusobna povezanost puta biosinteze ergosterola i sfingolipida je već dugi niz godina predmet istraživanja. Mnogobrojna novija istraživanja ukazuju na sinergijski učinak ergosterola i sfingolipida. U ovom radu provedena je analiza matematičkog modela biosinteze ergosterola pomoću programskih paketa WR Mathematica 8.0. i CellDesigner 4.3. U model biosinteze ergosterola uključene su reakcije puta biosinteze sfingolipida. Primjenom metoda lokalne i globalne osjetljivosti parametra, identificirani su ključni enzimi matematičkog modela. Rezultati lokalne analize uključuju transport ergosterola u staničnu membranu, uz hidrolize IPC-g i MIPC-g od strane enzima IPCaze, kao najosjetljivije reakcije modela. Kao ključni enzimi prilikom uspostavljanja ustaljenih vrijednosti, ističu se KDHS reduktaza, 4hidroksilaza i ceramid sintaza.

Abstract (english)

Interactions of ergosterol and sphingolipid biosynthesis pathways have been a subject of studies for many years. Numerous new studies indicate synergetic effects between ergosterol and sphingolipids. In this study a mathematical model analysis of ergosterol biosynthesis was performed, using WR Mathematica 8.0. and CellDesigner 4.3 software supports. Ergosterol biosynthesis model includes reaction rates of sphingolipid biosynthesis pathway. By applying local and global parameter sensitivity methods, critical enzymes of the model were identified. Local sensitivity analysis results include ergosterol transport to cellular membrane, along with hydrolosis of IPC-g i MIPC-g as the most sensitive reactions of the model. Upon establishing steady state values, the key enzymes are KDHS reductase, 4hydroxylase and ceramid synthase.
