Abstract

Primjena kultura stanica omogućava dokazivanje viralne replikacije mikroskopski, te godinama osigurava odgovarajući okoliš za detekciju i identifikaciju brojnih humanih viralnih patogena. Cilj ovog istraživanja bila je izolacija i umnažanje EV71 iz kliničkog uzorka cerebrospinalne tekućine bolesnika na RD staničnoj liniji, mikroskopska detekcija EV71 imunoflourescencijskom (IF) metodom i praćenjem citopatogenog učinka (CPE) virusa na RD stanicama, te određivanje titra umnoženog EV71 u mikrotitarskoj pločici određivanjem CPE virusa. Dobiveni rezultati pokazuju da EV71 postiže visoki titar (0,464-1 × 108/mL) kada se uzgaja na odgovarajućoj staničnoj liniji, kao što je RD stanična linija. Identifikacija EV71 potvrđena je primjenom imunoflourescencijske metode.

Abstract (english)

Cell cultures are convenient to examine microscopically for evidence of viral replication, and, for many years, have provided a desirable environment for the detection and identification of many human viral pathogens. The aim of this work was isolation and propagation of Enterovirus 71 from cerebrospinal fluid specimen in RD cell line, microscopic detection of the virus using immunofluorescence (IF) technique and cytopathic effect (CPE) as well viral titer determination by examining the occurrence of CPE in a RD cell monolayer. The obtained results have shown that EV71 reaches high titers (0,464-1 × 108/mL) when grown within appropriate cell line, such as RD cell line. Identification of EV71 was confirmed using the immunofluorescence method.
