Abstract

Razvoj stečene imunosti je kompleksan proces koji zahtjeva usku i kontroliranu suradnju različitih segmenata stečene i urođene imunosti. Dendritičke stanice imaju ključnu ulogu u povezivanju oba oblika imunosti tako što učinkovito prepoznaju različite strukturne molekularne uzorke (PAMP) uslijed čega dolazi do njihove aktivacije i naglašenog izlaganja MHC-antigen kompleksa na njihovoj površini. Aktivirane dendritičke stanice imaju sposobnost predočavanja i aktivacije naivnih limfocita T što dovodi do snažnog odgovarajućeg stečenog imunog odgovora nužnog za svladavanje patogena. Razvoj budućih cjepiva i terapeutika zahtjeva validirana in vitro testiranja koja uključuju sve ključne sudionike, pa tako i dendritičke stanice, a nužan preduvjet je razvoj učinkovitih protokola za dobivanje značajnog broja dendritičkih stanica. U ovom su radu istraživane in vitro diferencirane dendritičke stanice iz stanica koštane srži miša, te je potvrđeno da protokol rezultira zadovoljavajućim brojem stanica potencijalno sposobnih za sazrijevanje i predočavanje antigena.

Abstract (english)

Development of adaptive immunity is a complex process that requires tight and controlled cooperation of different adaptive and innate immune compartments. A central role for dendritic cells has been established since its discovery by Robert Steinman. Dendritic cells efficiently recognize different PAMPs and become activated along with emphasized MHCantigen presentation. Armed dendritic cells are capable to present and activate naive T cells leading to the established potent adaptive response necessary to combat pathogen. Development of the future vaccines and therapeutic approaches, require in vitro proof of concept testing using all key players, including dendritic cells. Development of efficient protocols to derive substantial numbers of dendtitic cells is therefore prerequisite. In our experiments we have tested deriving mDC in vitro from a murine bone marrow cells and confirmed that this protocol yields satisfactory cell number with the functional potential to mature and present antigens.
