Sažetak | Citotoksički učinak herbicida atrazina praćen je u staničnoj liniji bubrega 1 dan starih hrčaka (BHK 21 C13). BHK 21 C13 stanice rasle su u suspenziji na 37 °C u atmosferi 95% zraka i 5% CO2, u mediju za uzgoj Dulbecco's MEM uz dodatak 5% NCS-a (Newborn Calf Serum). Početna koncentracija stanica iznosila je 2x105 stanica/mL medija za uzgoj. Tijekom 72 sata metodom Trypan Blue praćen je citotoksički učinak atrazina u koncentraciji 20, 40, 60, 80, 100, 120 i 160 μg/mL medija za uzgoj. Pri koncentraciji atrazina od 80, 100, 120 i 160 μg/mL medija za uzgoj nakon 24 sata inhibicija rasta iznosila je od 34,19% do 54,19%. Nakon 48 sati tretmana atrazinom inhibicija rasta uočena je kod koncentracija od 60, 80, 100, 120 i 160 μg atrazina/mL medija za uzgoj i iznosila je od 23,39% do 65,27%. Nakon 72 sata tretmana atrazinom inhibicija rasta uočena je kod koncentracija od 40, 60, 80, 100, 120 i 160 μg atrazina/mL medija za uzgoj i iznosila je od 20,18% do 76,48%. IC50 vrijednost za atrazin za BHK 21 C13 stanice iznosila je 87,27 μg atrazina/mL medija za uzgoj nakon 72 sata. Iz dobivenih rezultata možemo zaključiti da koncentracija atrazina i vrijeme inkubacije utječu na proliferaciju BHK 21 C13 stanica. |
Sažetak (engleski) | Cytotoxic effect of herbicide atrazine was followed on cell line from one day old hamster kidney (BHK 21 C13 cell line). BHK 21 C13 cells were cultivated at 37°C in the atmosphere of 95% air and 5% CO2, in the Dulbecoo´s MEM medium with 5% NCS (Newborn Calf Serum). The initial concentration of cells was 2x105 cells/mL in cultivating medium. The cytotoxic effect of atrazine in concentrations of 20, 40, 60, 80, 100, 120 and 160 Ng/mL in cultivating medium was followed by Trypan Blue exclusion method during 72 hours. At atrazine concentrations of 80, 100, 120 and 160 μg/mL of cultivating medium after 24 hours growth inhibition ranged from 34.19 % to 54.19 %. After 48 hours of atrazine treatment, growth inhibition was detected at concentrations of 60, 80, 100, 120 and 160 μg of atrazine/mL of cultivating medium and it ranges from 23.39 % to 65.27 %. After 72 hours of atrazine treatement growth inhibition was detected at concentrations of 40, 60, 80, 100, 120 and 160 μg of atrazine/mL of cultivating medium and ranged from 20.18 % to 76.48 %. IC50 value for atrazine in BHK 21 C13 cell line was 87.27 μg of atrazine/mL of cultivating medium after 72 hours. From these results we can conclude that the concentration of atrazine and the time of incubation have effect on cell proliferation. |
