Određivanje kemokina IP-10i TARC u serumu djece s akutnom infekcijom respiratornim sincicijskim virusom

Abstract

Respiratorni sustav je neprestano izložen brojnim štetnim infekcijama i aerozagađenjima. U obrani domaćina od navedenih čimbenika sudjeluju mehanizmi specifične i nespecifične imunosti. Nespecifična imunost uključuje razne čimbenike, a jedan od njih su kemokini koje luče mnogobrojne stanice u organizmu. Kemokini su male molekule koje djeluju kemotaktički na upalne stanice i uzrokuju njihovu migraciju u upalna područja. Proizvodnja kemokina MIG (CXCL9) i IP-10 (CXCL10) kontrolirana je prisustvom citokina IFN-ỵ. Povišene koncentracije ovih kemokina se povezuju s virusnim infekcijama i diferencijacijom limfocita T u tip-1 smjeru. Tip-1 limfociti T na svojoj površini eksprimiraju CXCR3 kemokinski receptor koji specifično veže IP-10 i MIG te uzrokuje njihovu kemotaksiju važnu u obrani od virusa. Proizvodnju kemokina TARC (CCL17) i MDC (CCL22) moguće je potaknuti citokinom IL-4. Povišene koncentracije ovih kemokina se povezuju s alergijskim bolestima i diferencijacijom limfocita T u tip-2 smjeru. Tip-2 limfociti T na svojoj površini eksprimiraju CCR4 kemokinski receptor koji specifično veže MDC i TARC te uzrokuje njihovu kemotaksiju. U ovom istraživanju smo utvrdili povećanu koncentraciju kemokina IP-10 i TARC u serumima djece s akutnom infekcijom uzrokovanom respiratornim sincicijskim virusom, u odnosu na zdravu djecu dok nismo našli statističke razlike u koncetracijama kemokina MIG i MDC. Povišene koncentracije kemokina IP-10 i TARC potvrđuju sudjelovanje limfocita oba tipa imunoreakcija tijekom akutne infekcije te pojašnjava sličnosti patofiziologije virusne infekcije s astmatskim bolestima.

Abstract (english)

Respiratory tract represents a direct barrier between organism and environment, therefore constantly exposed to the numerous airborne pathogens and contaminants. Mucosal respiratory airway compartment employs different innate and adaptive immune mechanisms to recognize, neutralize and induce appropriate immune response against invading pathogen. Chemokines are heterogeneous small oligopeptide molecules involved in the chemotaxis of different cells expressing specific chemokine receptors. They are involved in the migration of leukocyte to the pathogen entrance, therefore necessary for an efficient immune reaction development. Besides their structural systematization, chemokines could be functionally grouped in-line with a specific type of immune response involvement. MIG (CXCL9) and IP-10 (CXCL10) chemokines contribute to activation and differentiation of type 1 T cells. Both chemokines are responsible for the chemotaxis of CXCR3-expressing type 1 cells capable to produce IFN-ỵ, which in turn could augment their levels in different viral infections. TARC (CCL17) and MDC (CCL22) chemokines contribute to development of type 2 immune response and higher levels could be found in allergic diseases. They regulate type 2 cell migration via specific CCR4 and their production is positively regulated by the IL-4. In this study, we have detected higher IP-10 and TARC levels in sera of RSV-infected infants. MIG and MCD levels were comparable with healthy infant levels. Increased IP-10 and TARC levels indicate that mixed type of adaptive immune response was acquired during acute RSV-infection which could contribute to immune related pathology observed in diseased infants.
