Abstract

Korištenjem GM biljki za ljudsku prehranu i/ili ishranu životinja javlja se potreba za kontroliranjem sigurnosti GM proizvoda. Velik broj metoda korištenih za analizu prisutnosti GMO-a od ranije je poznat, npr. PCR ili ELISA, no njihova primjena u GMO analizi i međusobno kombiniranje složen je proces. Razlozi su rastući broj novih GMO-a i/ili GM događaja, razvoj GMO tehnologije, nepoznanice vezane uz prijenos ili stabilnost nasljedne informacije te strukturu i funkcioniranje dijelova genoma. Svrha rada bila je ukazati na najvažnija ograničenja metoda, izazove koje za GMO analizu nosi daljnji razvoj GMO tehnologije te moguća rješenja koja se trenutno istražuju. Provedena je eksperimentalna kvalitativna analiza prisutnosti 35S-CaMV promotora u 20 uzoraka hrane pomoću klasičnog PCR-a i Real-time PCR-a. U 9 od 20 uzoraka detektirana je prisutnost 35S-CaMV promotora te je određen relativni udio (%) GM DNA soje u ukupnoj (tradicionalnoj i transgeničnoj) sojinoj DNA pojedinog poztivnog uzorka.

Abstract (english)

GM plants are used as human food and/or animal feed and it has become necessary to control the safety of GM products. A great number of methods used in the GMO analysis is already well known, e.g. PCR of ELISA, but the usage in GMO analysis and their combining is a very complex process. The reasons for this are growing number of new GMOs and/or GM events, development of GMO technology, unknown details regarding transfer or stability of genetic information and the structure and function of genome parts. The purpose of graduate thesis was to point the most important limitations and the challenges for GMO analysis methods brought by further development of GMO technology and possible solutions for them. qualitative analysis for the presence of 35S-CaMV promoter was experimentally conducted in 20 food/feed samples using traditional PCR and Real-Time PCR. In 9 of 20 samples the presence of 35S-CaMV promoter was detected and the relative amount (%) of GM soy DNA calculated in total soy (traditional and transgenic) DNA for every positive sample.
