Abstract

Svojstva koja imaju mnogi biološki aktivni spojevi nađeni u biljkama mogu se ispitati in vitro testovima citotoksičnosti pri čemu se često koriste humane stanične linije. Rezultati dobiveni određivanjem tzv. bazalne citotoksičnosti određuju učinak ispitivane tvari na postotak preživljenja stanica, a takav pristup je alternativa klasičnim in vivo pokusima na životinjama. U ovom radu, in vitro ispitivanje učinka subkritičnih vodenih ekstrakata peteljki jabuke i trešnje provedeno je na tumorskoj staničnoj liniji HeLa te je određena vijabilnost MTS metodom nakon 48 sati od tretmana. Djelovanje ekstrakata je uglavnom bilo inhibitorno na preživljavanje stanica. IC50 za ekstrakt trešnje iznosio je 5,8351% (v/v), a za ekstrakt jabuke 2,9547% (v/v). Blagi stimulacijski učinak zapažen je kod volumnog udjela ekstrakta trešnje od 5%. Rezultati dobiveni ovim istraživanjem ukazuju na potencijal primjene peteljki jabuke i trešnje kao izvor biološki aktivnih spojeva.

Abstract (english)

Properties of biologically active compounds found in plants can be tested with in vitro cytotoxicity tests using human cell lines. Results received by determining the so-called basal cytotoxicity determine the effect of the test substance on the percentage of cell survival, and this approach is an alternative to classical in vivo animal experiments. In this study, in vitro testing of the effects of subcritical water extracts of apple and cherry stems has been conducted on HeLa, human tumor cell line, and the viability was determined by MTS method after 72 hours of treatment. Results mostly showed inhibitory effects. IC50 for the cherry extract was 5,8351% (v/v), and for the apple extract 2,9547% (v/v). Mild stimulatory effect was noticed for cherry extract at volume ratio of 5%. These results indicate the potential of apple and cherry stems as a source of bioactive compounds.
