Abstract | U radu su ispitivani učinci lindana na koncentraciju glutationa u mozgu, testisu i prostati te na aktivnost kolinesteraza u serumu i mozgu štakora mužjaka prilagođenom metodom temeljenom na neenzimskoj reakciji s Ellmanovim reagensom. Utvrđena je linearnost metode u mjernom području od 3,12 do 200 nmol GSH, preciznost mjerenja unutar pokusa (%v = 0,67-1,75), točnost metode s iskorištenjem 99-103%. Dokazana je preciznost metode za određivanje aktivnosti kolinesteraza unutar pokusa (%v = 0,66-1,09), a točnost iste metode se potvrdila metodom promjene volumena seruma i citosola mozga u uzorku. Nakon sedmodnevnog tretmana lindanom (18 mg kg-1 tj. mase dnevno), a 24 h nakon posljednje doze, razina glutationa u tkivu mozga bila je 1,27 ± 0,212 nmol/mg tkiva, u tkivu testisa 3,67 ± 0,051 nmol/mg tkiva, a u tkivu prostate 0,74 ± 0,123 nmol/mg tkiva što nije statistički značajna promjena u odnosu na kontrolnu skupinu životinja (mozak: 1,08 ± 0,139 nmol/mg tkiva, testis: 3,52 ± 0,208 nmol/mg tkiva, prostata: 0,81 ± 0,103 nmol/mg tkiva). Lindan nije izazvao statistički značajne promjene u aktivnosti kolinesteraza u mozgu. U serumu je aktivnost kolinesteraza bila statistički značajno smanjena (0,40 ± 0,023 μmol min-1 mL-1 vs. 0,52 ± 0,014 μmol min-1 mL-1; p< 0,01). |
Abstract (english) | The effects of lindane on glutathione in brain, testis and prostate and on cholinesterase activities in serum and brain of adult male rat were tested by adjusted method established on nonenzymatic reaction with Ellman's reagent. Linearity of the method was confirmed by glutathione detection in the range of 3.12-200 nmol, and approved by within-assay precision (%v = 0.67-1.75 ) and accuracy of the method with recovery in range of 99-103 %. Within-assay precision (%v = 0.66- 1.09) of the method for cholinesterase activity determination was established and accuracy of the same method was confirmed by method of sample volume alteration. 24 hours after the last dose of 7-days treatment with lindane (18 mg kg-1 b. weight daily), and 24 h after the last dose the levels of reduced GSH were in brain 1.27 ± 0.212 nmol/mg tissue, in testis 3.67 ± 0.051 nmol/mg tissue, and in prostate 0.74 ± 0.123 nmol/mg tissue, which was not statisticaly significant alteration compared with control group of animals (brain: 1.08 ± 0.139 nmol/mg; testis: 3.52 ± 0.208 nmol/mg; prostate: 0.81 ± 0.103 nmol/mg). Applied doses of lindane did not induce significant alteration in cholinesterase activities in brain, but it caused statisticaly significant decrease (0.40 ± 0.023 μmol min-1 mL-1 vs. 0.52 ± 0.014 μmol min-1 mL-1; p< 0.01) in rat serum. |
