Abstract

Za uspješno odvijanje aerobnih bioprocesa nužno je koncentraciju otopljenog kisika održavati iznad kritične vrijednosti, a to se najlakše može izvesti promjenom parametara kao što su protok zraka i brzina rotacije mješala. U ovom radu dinamičkom metodom uz indirektno mjerenje odreñena je koncentracija otopljenog kisika odnosno volumni koeficijent brzine prijenosa kisika (kLa). Pri različitim kombinacijama procesnih parametara bioreaktora [brzina rotacije mješala (n= 200-400 min-1) i protoka zraka (F= 5-11 Lmin-1)] rezultati ispitivanja pokazuju da je optimalni protok zraka 10 L min-1. Povećanjem brzine rotacije mješala povećava se kLa vrijednost, stoga su najveće vrijednosti registrirane pri najvećoj brzini rotacije (400 min-1). U ovom istraživanju formiran je i regresijski model koji povezuje kLa (zavisna varijabla) s dvije nezavisne varijable [protok zraka (F) i brzina rotacije mješala (n)], te zadovoljavajuće opisuje promjene kLa u proučavanom intervalu procesnih parametara.

Abstract (english)

For succesful progress of an aerobic bioprocess it is necessary to keep the dissolved oxygen concentration above critical value, and this can be easily done by changing parameters such as air flow rate and stirrer rotation speed, respectively. In this work the concentration of dissolved oxygen as well as volumetric oxygen transfer rate coefficient (kLa) were estimated by dynamic method of indirect measurement. The influence of stirrer rotation speed ( n=200 – 400 min-1) and air flow rate (F= 5-11 Lmin-1) on kLa was investigated. The results of this investigation show the optimal air flow rate was 10 Lmin-1. By increasing stirrer rotation speed kLa value increase, therfore, the highest values observed at the highest stirrer rotation speed (400 min-1). In this work the regression model which corelate kLa (dependent variable) with two independent variables [ air flow (F) and stirring rate(n)], was established. This regression model apropriately describes the changes of kLa during investigation.
