Utjecaj ATPaze Sug1, podjedinice proteasoma i deubikvitinilaze Ubp8, podjedinice SAGA kompleksa na aktivaciju promotora PHO5 i PHO84 u kvascu Saccharomyces cerevisiae

Abstract

Proteasom je oligoproteinski kompleks koji je sa svojim proteolitičkim i neproteolitičkim funkcijama uključen u mnoge stanične procese, među kojima je i regulacija transkripcije. U ovom je radu ispitan utjecaj proteina Sug1 i Ubp8 na aktivaciju promotora PHO5 i PHO84 u kvascu Saccharomyces cerevisiae. Uloga proteina Sug1 je oslobađanje energije hidrolizom ATPa za razne funkcije proteasoma. Nađeno je da u stanicama mutanta kojima nedostaje taj protein dolazi do sporije aktivacije promotora PHO5 i PHO84 nego u stanicama divljeg tipa. Taj je utjecaj jače izražen kod promotora PHO5, nego PHO84. Protein Ubp8, podjedinica SAGA kompleksa, je enzim deubikvitinilaza. Funkcija tog enzima je deubikvitinilacija histona H2B. Rezultati su pokazali da je u stanicama mutanta ubp8 aktivacija promotora PHO5 i PHO84 sporija nego u stanicama divljeg tipa. Također je utvrđeno da stanice dvostrukog mutanta sug1- 25ubp8 pokazuju kumulativni efekt pri aktivaciji promotora PHO5, dok taj efekt nije utvrđen kod promotora PHO84.

Abstract (english)

The proteasome is a protein complex with various proteolitic and nonproteolitic functions involved in a number of cell process. One of them is regulation of transcription. In this paper the effects of Sug1 and Upb8 proteins on the PHO5 and PHO84 promoters in yeast Sacchromyces cerevisiae are shown. The role of the Sug1 protein, a subunit of the proteasome, is to hydrolyse ATP to relese energy, which is used to drive the specific functions of the proteasome. The activation of the PHO5 and PHO84 promoters is impaired in the mutant cells lacking Sug1 protein. This effect is stronger at the PHO5 than at the PHO84 promoter. Ubp8 protein is a subunit of SAGA complex. It’s role is to deubiquitylate the H2B histone. The results show that ubp8 mutants activate PHO5 and PHO84 promoters slower than the wild type. Also, it is shown that sug1-25ubp8 mutants display a sinthetic effect at the PHO5 promoter, but not at the PHO84 promoter.
