Abstract | Učinak α-tokoferola pri citotoksičkom učinku insekticida lindana praćen je u staničnoj liniji bubrega mladog hrčka (BHK 21 C13). BHK 21 C13 stanice rasle su u suspenziji na 37˚C u atmosferi 95% zraka i 5% CO2, u mediju za uzgoj BHK 21 medij (Glasgow MEM) uz dodatak 10% NCS-a (Newborn Calf Serum). Metodom Trypan Blue praćen je učinak α-tokoferola u koncentraciji 25 i 50 μg/mL medija za uzgoj na rast pri citotoksičkom učinku lindana u koncentracijama 20, 40, 60, 80 i 100 μg/mL medija za uzgoj tijekom 24 - 72 sata. Nakon 72 sata učinak vitamina E na dinamiku rasta stanica pri koncentraciji 25 μg/mL medija za uzgoj iznosi 109,33 %, a vitamina E u koncentraciji 50 μg/mL medija za uzgoj iznosi 115,18 % u odnosu na kontrolnu vrijednost. Pri koncentracijama lindana od 20 - 100 μg/mL medija za uzgoj inhibicija rasta iznosi 50,38 – 91,68 %, dok uz dodatak vitamina E 25 μg/mL medija za uzgoj iznosi 46,70 – 86,59 %, a uz dodatak vitamina E 50 μg/mL medija za uzgoj 30,50 – 91,51 %. IC50 vrijednosti za lindan u BHK 21 C13 stanicama u prisutnosti 25 μg/mL α-tokoferola povećane su za 4,8 %, a uz 50 μg/mL α-tokoferola 13 %. Znači prisutnost antioksidansa α-tokoferola u uvijetima praćenja citotoksičkog učinka lindana na BHK 21 C13 stanica ima protektivni učinak na preživljavanje stanica. |
Abstract (english) | In order to determine the effect of α-tocopherol during the cytotoxic effect of insecticide lindane, Baby Hamster Kidney (BHK 21 C13) cell line was used. BHK 21 C13 cells were cultivated at 37 OC in the atmosphere of 95% air and 5% CO2, in the medium BHK 21 (Glasgow MEM) with 10% NCS (Newborn Calf Serum). Effect of vitamin E in concentracions of 25 and 50 μg/mL of cultivating medium on cell growth during cytotoxic effect of lindane in concentrations of 20, 40, 60, 80 and 100 μg/mL of cultivating medium were determined during 24 - 72 hours by Trypan Blue colorimetric method. After 72 hours, effect of vitamin E on cell growth dynamics in concentration 25 μg/mL of cultivating medium was 109,33%, and by vitamin E in concentration 50 μg/mL of cultivating medium was 115,18%, compared to control value. At lindane concentrations of 20 - 100 μg/mL of cultivating medium, growth inhibitions were 50,38 – 91,68 %, and by the addition of vitamin E in concentration 25 μg/mL of cultivating medium, growth inhibitions were 46,70 – 86,59 %, and with the addition of vitamin E at concentration 50 μg/mL of cultivating medium, growth inhibitions were 30,50 – 91,51 %. IC50 for lindane in BHK 21 C13 cell line with the addition of vitamin E at concentration 25 μg/mL of cultivating medium was higher for 4,8 % and with the addition of vitamin E at concentration 50 μg/mL of cultivating medium for 13 %. It means that presence of an antioxidant α-tocopherol during the cytotoxic effect of lindane on BHK 21 C13 cell line have a protective effect. |
