Abstract

S-adenozil-L-metionin kemijski je spoj neophodan za život svake stanice. Njegova uloga je doniranje metilne skupine, u mnogim, za život neophodnim reakcijama. Važan je univerzalni metil donor. Nastaje iz metionina i ATP-a uz pomoć enzima metionin adenoziltransferaze. Tri metabolička puta važna su za ovu molekulu, to su: transmetilacija, trans-sulfuracija te biosinteza poliamina. Nedostatak SAMe-a pozantog kao S-adenozil-L-metionin u organizmu ili poremećaj u njegovoj homeostazi može dovesti do depresije, uništenja jetre, osteoporoze, alzheimerove bolesti i drugog. Pronađeno je da kvaščeve stanice hiperakumuliraju S-adenozil-L-metionin iz metionina u podlozi pod određenim uvjetima, koje se kasnije mogu koristiti kao dodatak prehrani. Cilj ovog rada je proučavanje utjecaja koncentracije D,L-metionina na rast kvasaca Saccharomyces cerevisiae TVG4, Saccharomyces uvarum LVK1, Candida utilis CUM te Kluyveromyces lactis KL71 i biosintezu SAMe, na glukoznim i laktoznim sintetskim podlogama.

Abstract (english)

S-Adenosyl-L-methionine is a chemical compound necessary for survival of all living cells. It is involved in a methyl group transfer in many reactions that are inevitable for life. It is very important as universal methyl donor. This substance comes from methionine and ATP, with a help of enzyme adenosyltransferase. There are three metabolic ways important for this molecule, i.e.: Transmethylation, transsulfuration and the polyamine biosynthesis. The lack of S-adenosyl methionine (SAMe) in an organism, or a disturbance in its homeostasis may cause of depression, liver damage, osteoporosis, Alzheimer disease, etc. It was found out, that under particular circumstances, the yeast cells hyperaccumulate S-Adenosyl-L-methionine from methonine as a base, and later on, it can be used as a food-additive. The purpose of this paper is to study the influence of the concentration of D, L-methonine to the growth of yeast biomass (Saccharomyces cerevisiae TVG4, Saccharomyces uvarum LVK1, Candida utilis CUM and Kluyveromyces lactis KL71), as well as to the biosynthesis of S-adenosyl methionine (SAMe).
