Abstract

Cilj ovog rada bio je ispitati utjecaj inhibitora HMG –CoA reduktaze na esteraze i leptin. Inhibitori HMG – CoA reduktaze ili statini su lijekovi koji se često koriste u liječenju hiperkolesterolemije. HMG – CoA reduktaza je enzim koji katalizira ključni korak u sintezi kolesterola: pretvorbu 3-hidroksi-3-metilglutaril-koenzima A (HMG-CoA) u mevalonat. Inhibitori HMG – CoA reduktaze su strukturni analozi HMG – CoA reduktaze, na čemu se i temelji njihov reverzibilni inhibicijski učinak na taj enzim. Leptin je hormon koji u najvećoj mjeri luči masno tkivo, a označava dovoljnu količinu energije, te je razina leptina u krvi veća kod ljudi čiji indeks tjelesne mase (ITM) je veći i koji imaju veći ukupini udio masti. Pretilost, dakle, uzrokuje povišene razine leptina u krvi, ali i povišene razine kolesterola i ukupnih lipida. Smanjenjem tjelesne mase postiže se poboljšanje profila lipida u krvi, a jedan od načina za mršavljenje je prehrambena intervencija uz odgovarajuću tjelesnu aktivnost.

Abstract (english)

The aim of this final thesis was to examine how HMG-CoA reductase inhibitors act on esterases and leptin. HMG-CoA reductase inhibitors, also known as statins, are class of lipid-lowering medications, that are often used in the treatment of hypercholesterolemia. HMG - CoA reductase is the enzyme that catalyzes a key step in the synthesis of cholesterol: the conversion of 3-hydroxy-3-methylglutaryl-coenzyme A ( HMG- CoA ) to mevalonate. HMG - CoA reductase inhibitors are structural analogs of HMG - CoA reductase , on which is based their reversible inhibitory effect on that enzyme. Leptin is a hormone made by adipose cells that helps to regulate energy balance by inhibiting hunger. Level of leptin in the blood is higher in people whose body mass index (BMI) is higher. Obesity, therefore, causes increased levels of leptin in blood and also higher levels of cholesterol and total lipids. Improvement of the lipid profile in blood can be achieved by reducing body weight, and one way to lose weight is a nutritional intervention with appropriate physical activity.
