Abstract

Crijevna mikrobiota, iako relativno novo područje istraživanja, je pobudila snažan interes za istraživanjem uslijed svojeg velikog utjecaja na zdravlje i dobrobit ljudi. Zbog raznih etičkih ograničenja pri istraživanju crijevne mikrobiote, razvijaju se razni in vitro sustavi kojima je cilj što bolje simulirati uvjete u crijevu čovjeka. Postoji više ograničavajućih faktora pri istraživanju mikrobiote u in vitro sustavima od kojih je najznačajniji nemogućnost repliciranja svih fizioloških uvjeta prisutnih u organizmu čovjeka. Svrha ovog rada je bila ispitati koliko je odabrani i prilagođeni in vitro sustav podoban za održavanje postojanog sastava inokulirane mikrobiote, pratiti odgovor mikrobiote na uvjete uzgoja u in vitro sustavu, utjecaj hranjive podloge na mikrobiotu te reproducibilnost sustava. Nakon provedenog procesa, utvrđeno je da je korišteni in vitro sustav reproducibilan i dobro simulira uvjete debelog crijeva te je za buduća istraživanja potrebno optimizirati sastav hranjive podloge kako bi se održala bioraznolikost mikrobiote što sličnija onoj iz početnog inokuluma.

Abstract (english)

Gut microbiota, although relatively new research area, has aroused a strong interest in researching community due to its great impact on the human health and wellbeing. Because of various ethical norms in the research of gut microbiota, new in vitro systems are being developed. Their goal is to simulate the physiological conditions in the human gut. There are a few limiting factors in researching the gut micorbiota in in vitro systems and the biggest one is its inability to replicate all the conditions that exist in vivo in the colon. The aim of this thesis was to evaluate how adequate is the chosen and adapted in vitro system to mantain a stable composition of the inoculated micorbiota, to keep track of the responses that the microbiota has to the conditions in the system, to investigate the impact of SIEM on the microbiota composition and the reproducibility od the in vitro system. The experiment has given the results that the used in vitro system is reproducible and is able to adequately simulate the conditions present in the human colon. In the future experiments, it is necessary to optimize the composition of SIEM to be able to mantain the biodiversity of micorbiota and the composition as similar as possible to the original inoculate.
