Abstract

Eutektička otapala pripadaju novoj generaciji nehlapljivih i stabilnih otapala te se intenzivno proučavaju kao zamjena za organska otapala u kemijskoj i biotehnološkoj industriji. U ovom radu ispitana je mogućnost primjene prirodnih eutektičkih otapala na bazi kolin-klorida u enantioselektivnoj hidrolizi (R, S)-1-feniletil-acetata uz lipazu B kao biokatalizator. Prirodna eutektička otapala pokazala su se povoljnijim otapalima od konvencionalnih otapala (organska otapala i pufer) za ispitivanu hidrolizu obzirom na ispitane parametre (ɳ, ee, VP i VE), pri čemu se otapalo ChGlc50 % pokazalo kao najbolje. Optimiranjem reakcijskih uvjeta u ChGlc50% pronađeni su optimalni uvjeti za hidrolizu koji se odnose na omjer enzima i supstrata, temperaturu i broj okretaja. Također, ispitan je utjecaj ultrazvuka i mikrovalova, bilo da su korišteni istovremeno ili samostalno, na volumetrijsku produktivnost enantioselektivne hidrolize (R, S)-1-feniletil-acetata pri čemu se najveće povećanje produktivnosti postiglo istovremenom upotrebom ultrazvuka i mikrovalova.

Abstract (english)

Natural eutectic solvents belong to the new generation of non-volatile and stabile solvents, and are intensively studied as a replacement for organic solvents in chemical and biotechnological industry. Within this work the possibility of application cholinum-based natural deep eutectic solvents for enantioselective hydrolysis of (R,S)-1-phenylethyl acetate with lipase B as biocatalyst was studied. All enantioselective reactions in natural deep eutectic solvents were successfully conducted, and those solvents were superior to organic solvents according to all tested parameters (ɳ, ee, VP and VE) where solvent ChGlc50% was found to be the best between all used. By optimizing the reaction conditions within the solvent ChGlc50% the most optimal conditions have been shown and are refer to ratio of enzyme and substrate, temperature and number of turnovers. The impact of ultrasound (US) and microwave irradiation (MW), either used independently or simultaneously, on volumetric productivity of enantioselective hydrolysis of (R,S)-1-phenylethyl acetate, was also examined and the highest productivity was achieved by simultaneous use of US and MW.
