Abstract

Zbog svakodnevne primjene veterinarskih lijekova obavezna je kontrola hrane životinjskog podrijetla na rezidue u hrani. Cilj ovog rada bio je validirati ELISA metodu za određivanje sulfonamida i kloramfenikola u uzorcima različitih mesnih proizvoda (pašteta, buđola, mesni doručak, zimska salama, dimljena šunka). Ključni validacijski parametri bili su: selektivnost/specifičnost i sposobnost detekcije, iskorištenje, ponovljivost i preciznost, granica detekcije, granica kvantifikacije i robusnost. Koncentracije sulfonamida i kloramfenikola bile su niže od najviše dopuštene koncentracije, odnosno granice najmanje zahtijevane učinkovitosti izvedbe metode prema čemu je izračunata sposobnost detekcije i potvrđena selektivnost/specifičnost. Iskorištenje, ponovljivost i preciznost također su se nalazile unutar granica određenih zakonom. Granice detekcije i kvantifikacije za sulfonamide iznosile su 0,627 µg kg-1 i 2,891 µg kg-1, a za kloramfenikol 0,019 µg kg-1 i 0,068 µg kg-1. Dokazana je robusnost metode za određivanje kloramfenikola. S obzirom na dobivene rezultate i postavljene kriterije prihvatljivosti može se zaključiti da je validirana ELISA metoda prikladna za dokazivanje sulfonamida i kloramfenikola u mesnim proizvodima.

Abstract (english)

Everyday use of veterinary drugs makes it necessary to control the residues in food. That is why it is necessary to control animal source foods. The aim of this paper was to validate the ELISA method for determining sulphonamides and chloramphenicol in different processed meat products (pate, smoked neck meat, meat breakfast, winter salami and smoked ham). Key validation parameters were: selectivity/specificity and detection capability, recovery, repeatability and precision, limit of detection and quantification, and robustness. Sulphonamides and chloramphenicol concentrations were lower than maximum residue limit and minimum required performance limit of method, respectively, thereby calculating the detection capability and confirmed selectivity/specificity. The recovery, repeatability and precision were also within the limits set by regulations. The limits of detection and quantification for sulphonamides were 0,627 µg kg-1 and 2,891 µg kg-1, respectively, and for chloramphenicol 0,019 µg kg-1 and 0,068 µg kg-1, respectively. Method for chloramphenicol determination showed to be robust. The results of this study confirmed suitability of validated ELISA method for determination of sulphonamides and chloramphenicol in meat products.
