Abstract

Proveden je saržni uzgoj gljive T. versicolor u tekućoj hranjivoj podlozi s ciljem
optimiziranja proizvodnje ekstracelularnih lignocelulolitičkih enzima. Uzgoj se provodio u pet različitih
hranjivih podloga sa tri različita izvora ugljika: glukoza, lipa i hrast. Izvori ugljika bili su sami u podlozi
ili u kombinaciji s glukozom u ekvivalentnom masenom odnosu (lipa i glukoza; hrast i glukoza). Tijek
biosinteze enzima pratio se mjerenjem enzimske aktivnosti lakaza, mangan peroksidaza i celobioza
dehidrogenaza metodom mjerenja promjene apsorbancije o vremenu za određeni supstrat pri
određenoj valnoj duljini. Hemicelulazna i celulazna aktivnost mjerila se u 18. danu uzgoja DNSA
metodom. Hrast se pokazao kao dobar induktor sinteze lignolitičkih i hemicelulolitičkih enzima.
Najoptimalnija hranjiva podloga za proizvodnju ekstracelularnih enzima bila je podloga sa
kombiniranim izvorom ugljika, hrastom i glukozom, u kojoj je u 18. danu uzgoja dobivena lakazna
aktivnost iznosila 0,658 U/mL, mangan peroksidazna aktivnosti iznosila 0,073 U/mL, ksilanazna
aktivnost iznosila 1,635 U/mL, pektinazna aktivnost iznosila je 0,503 U/mL i arabinazna aktivnost
iznosila je 0,689 U/mL.

Abstract (english)

Batch cultivation of fungus T. versicolor was conducted in the liquid growth medium in order to optimise the production of extracellular lignocelulolytic enzymes. Cultivation was conducted in five different growth medium with three different carbon sources: glucose, lime tree and oak. Carbon sources were either the only carbon source or in combination with glucose in equivalent mass ratio (lime and glucose; oak and glucose). The course of enzyme biosynthesis was monitored by measuring the enzyme activity of laccases, manganese peroxidases and cellobiose dehydrogenases by measuring the slope of the absorption of specified substrate at a specified wavelength. Hemicellulase and cellulase activity was measured during the 18th day of cultivation using the DNSA assay. Oak was shown to be a good inductor of ligninolytic and hemicellulolytic enzyme synthesis. The most optimal growth medium was the medium with the combined carbon sources, oak and glucose, at which during the 18th day of cultivation the obtained laccase activity was 0.658 U/ml, manganese peroxidase activity was 0.073 U/ml, xylanase activity was 1.635 U/ml, pectinase activity was 0.503 U/ml and arabinase activity was 0.689 U/ml.
