Abstract

Posljednjih nekoliko desetljeća pokušavaju se pronaći alternative korištenju seruma animalnog podrijetla u tehnologiji životinjskih stanica zbog određenih nedostataka poput pojave mogućih kontaminacija i povećanih troškova procesa pročišćavanja. Biljni proteinski hidrolizati koriste se stoga kao djelomična ili potpuna zamjena za serum budući da sadrže mnoge hranjive tvari i komponente nužne za uspješan rast i proliferaciju kulture životinjskih stanica. Cilj ovog rada je istražiti i ispitati učinak proteinskog hidrolizata uljne pogače lana dobivenog pomoću tri enzima: Alcalase, Neutrase i Protamex na rast i produktivnost CHO-DP 12 stanične linije u kemijski definiranom mediju bez seruma. Ispitani su učinci hidrolizata dodanih u medij u dvije različite koncentracije: 2 g L-1 i 0.5 g L-1. Istraživanje pokazuje da dodatak hidrolizata dobivenih Protamexom i Neutrasom u koncentraciji od 2 i 0,5 g L-1 povoljno utječe na rast, ali i na produktivnost CHO-DP12 stanica u odnosu na stanice uzgajane u mediju bez dodatka hidrolizata. Hidrolizati dobiveni hidrolizom pomoću enzima Alcalase, dodani u obje ispitivane koncentracije, uzrokuju smrt stanične linije.

Abstract (english)

Numerous researches have beed done during the last few decades in order to find alternatives for using the animal origin serum due to several deficits like possible contaminations or additional costs for purification. Plant derived protein hydrolysates are therefore used as a partial or complete substitute because of containing various nutriments and similar components needed for a successful growth and proliferation of an animal cell culture. The aim of this paper is to find and test the effect of the protein hydrolysate of flaxseed oil cake obtained by three enzymes, Alcalase, Neutrase and Protamex on cell growth and productivity of CHO DP-12 cell line in a chemically defined medium without serum. Moreover, the effect of the examined hydrolysate in media has been analysed in two different concentrations: 2 g L-1 and 0,5 g L-1. This research shows that the addition of two hydrolysates, which are obtained by Protamex and Neutrase in the 2 and 0,5 g L-1 concentration, has a positive impact on the growth and productivity of CHO-DP12 cells compared to the cells in a hydrolysate-free medium. Nevertheless, the hydrolysates obtained by Alcalase causes the starvation of cell lines in both examined concentrations.
