Abstract

Iskazivanju funkcionalnih učinaka probiotika u gastrointestinalnom traktu (GIT), može doprinijeti i primjena različitih prebiotika koji selektivno potiču rast poželjnih bakterija in situ. Stoga je cilj ovog rada bio istražiti dodatak 3 različita prebiotika: inulina s fruktooligosaharidima, manitola i laktuloze na učinkovitost mikroinkapsulacije bakterijskih stanica soja Lactobacillus fermentum D12 u alginatu. Lb. fermentum D12 je autohtona bakterija mliječne kiseline koja se istražuje kao potencijalna probiotička starter kultura, stoga je za biotehnološku proizvodnju ovog soja važno i definirati tehnološka svojstva primjerice preživljavanje tijekom procesa mikroinkapsulacije i liofilizacije. Nastavno je u ovom radu provedena i liofilizacija mikroinkapsuliranih bakterijskih stanica soja D12 uz dodatak obranog mlijeka kao lioprotektora. Prema rezultatima, dodatak prebiotika tijekom mikroinkapsulacije bakterijskih stanica D12 u alginatu ima povoljan učinak na preživljavanje Lb. fermentum D12, što proizlazi iz većeg broja preživjelih bakterijskih stanica u usporedbi s kontrolom. Prisutnost čiste kulture bakterijskog soja D12 u mikroinkapsuliranim i liofiliziranim pripravcima potvrđena je RAPD metodom. U konačnici, liofilizirane i mikroinkapsulirane stanice soja Lb. fermentum D12 preživljavaju u visokom broju (CFU/ml) simulirane uvjete GIT, što ukazuje na zaštitni učinak primjenjenih procesa čime se ujedno doprinosi funkcionalnosti probiotičkog soja in vivo.

Abstract (english)

The application of various prebiotics, that selectively promotes the growth of desired bacteria in situ, may also contribute to the functional properties of probiotics in the gastrointestinal tract (GIT). Therefore, the aim of this thesis was to determine the influences of 3 different prebiotics: inulin with fructo-oligosaccharides, mannitol and lactulose on the efficiency of the microencapsulation of Lb. fermentum D12 bacterial cells in alginate. Lb. fermentum D12 is an autochthonous lactic acid bacterium that has been explored as a potential probiotic starter culture, so for the biotechnological production of this strain, it is important to define its technological properties, such as survival during the microencapsulation and lyophilization. In this work, the lyophilization of microencapsulated bacterial cells of strain D12, with the addition of skim milk as a lioprotector, was also carried out. According to the results, the addition of prebiotics during the microencapsulation of bacterial cells D12 in the alginate has a positive effect on the survival of Lb. fermentum D12, resulting from a higher number viable bacterial cells compared to the control. The presence of pure culture of bacterial strain D12 in microencapsulated and lyophilized formulations was confirmed by the RAPD method. Finally, lyophilized and microencapsulated Lb. fermentum D12 cells survive in high viable cell counts (CFU/ml) in simulated conditions of the GIT, what implies on the protective effect of the applied processes, thereby also contributing to the functionality of the probiotic strain in vivo.
