Karakterizacija divljih tipova virusa izoliranih iz zaraženih animalnih tkiva u svrhu proizvodnje virusnih cjepiva

Abstract

Virusne infekcije predstavljaju veliki zdravstveni i ekonomski problem u svijetu, a kako bi se smanjile posljedice takvih infekcija mnoge su farmaceutske tvrtke svoju proizvodnju usmjerile na proizvodnju cjepiva kao jedini način borbe protiv virusa. Međutim, veliki izazov u razvoju budućih generacija virusnih cjepiva predstavljaju brze mutacije virusa i izmjena genetskog materijala. Stoga je u svrhu razvoja i proizvodnje virusnih cjepiva, te stvaranja banke virusa, cilj rada bio iz zaraženih animalnih tkiva izolirati divlji tip virusa Bovine herpesvirus 1, Bovine parainfluenza 3 i Bovine rotavirus A, te je qRT-PCR metodom uz primjenu odgovarajućih primera potvrđeno kako su izolirane virusne čestice upravo željeni virusi. Također je proveden Test sterilnosti kojim je dokazano kako banke virusa ne sadrže kontaminante s obzirom da nije došlo do porasta kolonija na TSA hranjivim podlogama, dok je Test na mikoplazme pokazao odsutnost mikoplazmi u bankama virusa.

Abstract (english)

Viral infections are a major health and economic world's problem. In order to reduce the consequences of viral infections, many pharmaceutical companies have focused their production on the production of vaccines as the only way to overcome the virus. However, a major challenge in the development of future generations of viral vaccines is the rapid mutations of viruses and changes in genetic material. Therefore, for the intent of development and production of viral vaccines and creation of a virus bank, the purpose of the study was to isolate wild type of virus Bovine herpesvirus 1, Bovine parainfluenza 3 and Bovine rotavirus A from infected animal tissues and it was confirmed by qRT-PCR that the isolated viral particles are the desired viruses. There is also performed a Sterility test which proved that the virus banks did not contain contaminants because there was no colony increase on TSA nutrient media and Mycoplasma test which showed the absence of mycoplasmas in the virus banks.
