Učinak hidrolizata proteina uljne pogače sjemenki lana na rast, produktivnost i metabolizam CHO DP-12 stanične linije

Abstract

Životinjski serum je standardni dodatak mediju za uzgoj životinjskih stanica. Međutim, zbog određenih nedostataka već više od tri desetljeća istražuje se upotreba biljnih hidrolizata proteina kao zamjene serumu u biotehnološkim postupcima proizvodnje rekombinantnih proteina pomoću životinjskih stanica. Cilj ovog rada bio je utvrditi utjecaj hidrolizata proteina uljne pogače sjemenki lana, dobivenih pomoću proteolitičkih enzima Alcalase, Neutrase i Protamex, na rast, produktivnost i metabolizam CHO DP-12 stanične linije. Osim utjecaja hidrolizata ukupnog sadržaja peptida, ispitan je i utjecaj peptidnih frakcija veličine do 10 i do 1 kDa kao dodataka mediju za uzgoj u koncentraciji od 2 i 0.5 g Lˉ¹ . Rezultati su pokazali kako dodani hidrolizati i njihove frakcije nemaju značajan utjecaj na rast stanica, dok je pokazan pozitivan utjecaj na produktivnost. Dodatak hidrolizata dobivenog pomoću enzima Protamex u koncentraciji 2 g Lˉ¹ rezultirao je visokom produktivnošću dok je istovremeno usporio rast stanica.

Abstract (english)

Animal serum is a standard media supplement for animal cultures, but because of its disadvantages, for more than three decades, the use of plant protein hydrolysates as a serum replacement in biotechnological processes for the production of recombinant proteins with cell cultures is researched. This study aimed to determine the influence of flaxseed oil cake protein hydrolysates, obtained by proteolytic enzymes, Alcalase, Neutrase and Protamex, on growth, productivity and metabolism of CHO DP-12 cell line. In addition to the influence of hydrolysates of the total peptide content, fractions of peptides size up to 10 kDa and 1 kDa were tested as culture media supplements in concentrations of 2 and 0.5 g L-1. Research showed that the added hydrolysates and their fractions did not have a significant impact on cell growth while a positive effect on productivity was shown. The addition of hydrolysate obtained with Protamex in a concentration of 2 g Lˉ¹ resulted in high productivity while at the same time slowing down cell growth.
