Abstract

Ekstrakt dobričice se već stoljećima upotrebljava u narodnoj medicini za ublažavanje i liječenje raznih zdravstvenih tegoba. Glavne farmakološki aktivne tvari ekstrakta dobričice čine polifenolni spojevi, koje karakterizira izniman antioksidacijski i antimikrobni potencijal, čime pružaju zaštitu ljudskom organizmu od bakterijskih infekcija, kao i od razvoja kroničnih bolesti povezanih s oksidativnim stresom. Cilj ovog rada je ispitati biološki učinak ekstrakta dobričice na stanične makromolekule, odabrane tumorske stanične linije epitela probavnog sustava, kao i na glavne predstavnike humane mikrobiote te utjecaj ekstrakta na njihovo vezanje za epitelne stanice gastrointestinalnog trakta. Dobiveni rezultati pokazali su da citotoksičnost ekstrakta ovisi o vrsti tretiranih stanica te da se prooksidativni učinak uglavnom smanjuje s produljenjem vremena izloženosti stanica. Utvrđen je i protektivni učinak ekstrakta od oksidacijskih oštećenja induciranih hidroksilnim radikalima na genetički materijal. Biološki aktivni spojevi prisutni u ekstraktu dobričice pokazali su snažnu antimikrobnu aktivnost prema patogenim bakterijama E. coli i S. aureus te sposobnost da dovedu do promjena u adheziji bakterija za epitelne stanice gastrointestinalnog trakta.

Abstract (english)

The ground ivy extract has been used for centuries in folk medicine to alleviate and treat various health ailments. The main pharmacologically active substances of ground ivy extract are polyphenolic compounds, which are characterized by exceptional antioxidant and antimicrobial potential, thus providing protection to the human body from bacterial infections as well as from the development of chronic diseases associated with oxidative stress. The aim of this study is to examine the biological effect of ground ivy extract on cellular macromolecules, selected tumor cell lines of the digestive epithelium, as well as on the representatives of the human microbiota and the influence of the extract on their adherence to gastrointestinal epithelial cells. The obtained results showed that the cytotoxicity of the extract depends on the type of treated cells and the prooxidative effect generally decreases with prolongation of cell exposure time. The protective effect of the extract from oxidative damages induced by hydroxyl radicals on genetic material was also determined. Biologically active compounds present in the ground ivy extract showed strong antimicrobial activity against pathogenic bacteria E. coli and S. aureus and the ability to change bacterial adhesion to epithelial cells of the gastrointestinal tract.
