Abstract

U ovom radu istraživana je sposobnost bakterije Paenibacillus polymyxa DSM 742 da proizvede enzime za razgradnju lignocelulozne biomase. Lignoceluloza je osnovni gradivni materijal biljaka, koji se može iskoristiti za proizvodnju biogoriva, biokemikalija i biopolimera, no mora se prethodno razgraditi na gradivne elemente. U ovom radu proveden je uzgoj bakterije Paenibacillus polymyxa DSM 742 u kiselom i bazičnom pH na pet različitih izvora ugljika kako bi se ispitala njena sposobnost proizvodnje lignocelulotičkih enzima. Pratio se rast biomase, ukupna koncentracija proteina i enzimska aktivnost tijekom 25 h uzgoja u tikvicama i bioreaktoru s miješalom. Najveća specifična enzimska aktivnost izmjerena je na podlogama sa fruktozom kao izvorom ugljika u kiselom pH. Maksimalna specifična enzimska aktivnost ksilanaze iznosila je 0,7 U/mg, egzoglukanaze 1,8 U/mg i endoglukanaze 1,5 U/mg. U biorektoru uzgoj je provoden fruktozi u kiselom pH i dobivena je maksimalna enzimska aktivnost ksilanaze 9,3 U/mg, aktivnosti egzo- i endoglukanaza su bile manje od 0,8 U/mg.

Abstract (english)

The aim of this study was to investigate the ability of the bacterium Paenibacillus polymyxa DSM 742 to produce enzymes for the degradation of lignocellulosic biomass. Lignocellulose is the basic building material of plants that can be used for the production of biofuels, biochemicals and biopolymers, but it must first be broken down into building elements. In this study, the cultivation of the bacterium Paenibacillus polymyxa DSM 742 was carried out in acidic and basic pH on five different carbon sources to test its ability to produce lignocellulosic enzymes. Biomass growth, total protein concentration and enzyme activity were monitored during 25 h of cultivation in flasks and a stirred tank bioreactor. The highest specific enzyme activity was measured on substrates with fructose as a carbon source in acidic pH. The maximum specific enzyme activity of xylanase was 0.7 U/mg, exoglucanase 1.8 U/mg and endoglucanase 1.5 U/mg. In the bioreactor, cultivation was carried out on fructose in an acidic pH and the maximum enzymatic activity of xylanase was 9.3 U/mg, the activities of exo- and endoglucanase were less than 0.8 U/mg.
