Abstract

Do danas su dokumentirani višestruki korisni učinci probiotika na zdravlje domaćina. Jedan od glavnih kriterija za izbor probiotičkog soja je preživljavanje u gastrointestinalnom sustavu te adhezija na crijevni epitel kako bi se na ciljanom mjestu iskazali korisni učinci. Obzirom da je nužno da probiotici u što većem broju prežive stresne uvjete, često se koriste tehnike koje štite stanice od nepovoljnih uvjeta. Primjer primjene takvih matriksa je mikroinkapsulacija u alginatu i želatini. Također, pripravljenim proizvodima važno je maksimalno produljiti trajnost pa se koriste tehnike liofilizacije koje omogućavaju dulje skladištenje. U ovom radu je, optimiranjem tehnika mikroinkapsulacije, odabran najučinkovitiji način zaštite odabranog probiotičkog soja Lactiplantibacillus plantarum MB18. Dokazana je visoka učinkovitost mikroinkapsulacije za svaki omjer korištenih matriksa. Alginat i želatina bili su jednako prikladni nosači probiotičkog soja povećavajući mu preživljavanje u in vitro uvjetima želučanog soka i tankog crijeva. Kod svih mikroformulacija dokazana je uspješna fermentacijska aktivnost soja L. plantarum MB18, zbog čega se pripravljene liofilizirane mikrokapsule mogu koristiti kao potencijalne probiotičke funkcionalne starter kulture.

Abstract (english)

To date, numerous beneficial effects of probiotics on host health have been documented. One of the main criteria for the selection of a probiotic strain is its survival in the gastrointestinal tract and its adhesion in the intestinal epithelium to exert beneficial effects at the target site. Since it is necessary for probiotics to survive the stressful conditions in as large number as possible, various techniques are often used to protect cells from adverse conditions. An example of the application of such matrices is microencapsulation in alginate or gelatin carriers. In addition, it is important to extend the shelf life of the prepared products as much as possible, so freeze-drying techniques are used to allow longer storage period. In this work, by optimizing microencapsulation techniques, the most effective method for protecting the selected probiotic strain Lactiplantibacillus plantarum MB18 was chosen. The high efficiency of microencapsulation was demonstrated for each ratio of matrices used. Alginate and gelatin were equally suitable as carriers for the probiotic strain and increased its survival under in vitro conditions in gastric juice and small intestine. Successful fermentation activity of L. plantarum MB18 strain was demonstrated in all microformulations, therefore the produced lyophilized microcapsules can be used as potential probiotic functional starter cultures.
