Abstract

Negativni učinci na okoliš i nestašica fosilnih goriva dovode do sve veće potražnje za alternativnim izvorima goriva i energije. Oleaginozni kvasci imaju sposobnost nakupljanja lipida koji se mogu primijeniti u proizvodnji biodizela kao alternativa naftnom dizelu. Cilj ovog rada bio je istražiti učinak osmotskog stresa na stanicu kvasca Rhodotorula sp. MG2 izazvanog dodatkom natrijeva klorida u koncentraciji od 25 do 75 g/L. Učinak stresa na stanicu praćen je određivanjem rasta i sastava stanice te udjela karotenoida i lipida. Povećanjem koncentracije natrijevog klorida u hranjivoj podlozi raste udjel lipida, proteina, karotenoida te anorganskih tvari u stanici. Najveća koncentracija karotenoida u ekstraktu od 50,40 mg/L proizvedena je dodatkom najveće koncentracije natrijeva klorida od 75 g/L. Također, najveći udio lipida u stanici od 41,3 % izmjeren je kod najveće koncentracije natrijeva klorida. Metanolski esktrakt stanične biomase te egzopolisaharidi kvasca Rhodotorula sp. MG2 nisu pokazali antibakterijski i antimikotski učinak. Kvasac također ne proizvodi soforolipide.

Abstract (english)

The negative effects on the environment and the shortage of fossil fuels lead to an increasing demand for alternative sources of fuel and energy. Oleaginous yeasts produce lipids that can be used in the production of biodiesel as an alternative to petroleum diesel. The aim of this work was to investigate the effect of osmotic stress on the yeast cell Rhodotorula sp. MG2 induced by the addition of sodium chloride in concentration in the range from 25 to 75 g/L. The effect of stress on the cell was monitored by determining cell growth and composition, as well as the proportion of lipids, proteins, carotenoids and inorganic substances in the cell. The highest concentration of carotenoids in the extract of 50,40 mg/L was produced by the addition of the highest concentration of sodium chloride of 75 g/L. Also, the highest content of lipids in the cell of 41,3 % was determined at the highest concentration of sodium chloride. Methanolic extract of cellular biomass and exopolysaccharides of the yeast Rhodotorula sp. MG2 did not show antibacterial and antifungal effects. Yeast also does not produce sophorolipids.
