Abstract

Euglena gracilis je jednostanični protist koji se giba na različite načine izazvane svjetlom; fototaksijom (ciljani pokret prema ili od izvora svjetlosti) i fotofobijom, odnosno naglim zaokretom zbog jakog povećanja (step-up) ili smanjenja (step-down) intenziteta svjetlosti. Negativna i pozitivna fototaksija je proučavana dodatkom različitih koncentracija Ca2+ i Cr6+ iona (0.5, 1.0 i 2.0 mM) u podlogu za uzgoj. Pokretljivost i kutovi fototaksijskog gibanja E. gracilis tijekom 14 dana uzgoja su snimani u obliku video filma (30 sekundi) putem kamere postavljene na okular mikroskopa. Niske koncentracije Ca2+ i Cr6+ iona usporile su promjenu smjera gibanja kao posljedicu negativne fototaksije, dok je tijekom pozitivne fototaksije reorjentacija E. gracilis bila pojačana.

Abstract (english)

Euglena gracilis, a unicellular freshwater protist exhibits different photo movement responses, such as phototaxis (oriented movement toward or away from the light source) and photophobic (abrupt turn in response to a rapid increase (step-up) or decrease (step-down) in the light influence rate) responses. Negative and positive phototaxis was studied at various external Ca2+ and Cr6+ concentrations (0.5, 1.0 and 2.0 mM). The swimming velocity and the angles of phototactic orientation were measured with individual cells after different times of incubation in the Ca2+ and Cr6+ solutions (1-14 days). The movements of E. gracilis are videotaped (30 sec) by means of camera mounted on a microscope. Low Ca2+ and Cr6+ concentrations slow down the reorientation during negative phototaxis, whereas during positive phototaxis reorientation of E. gracilis is enhanced.
