Abstract | Kvasac Trichosporon oleaginousus primijenjen je u procesu istovremenog ošećerenja i proizvodnje lipida (SSF) s predobrađenim kukuruznim oklascima uz omjer tekućeg i čvrstog dijela podloge (LSR) 20:1-8:1 mL g-1. Nakon 144 sata šaržnog uzgoja, produktivnost nakupljanja lipida povećavala se s povećanjem omjera LSR do maksimalne vrijednosti od 1,69 g L-1 d-1 uz omjer LSR od 8:1 mLg-1. Najveća koncentracija lipida od 11,5 g L-1 postignuta je uz omjer LSR od 8:1 mL g-1 nakon 312 sati uzgoja, dok je najveći udjel lipida 172,8 mg g-1 taloga koji je sadržavao nehidrolizirani supstrat i biomasu kvasca postignut uz LSR of 10:1 mL g-1 nakon 240 h. Nadalje, proveden je šaržni SSF uz prihranu supstratom u obrocima po 2,5 i 5 g. Šaržnom prihranom postignut je ukupni omjer LSR od 5:1 i 6,7:1 mL g-1. Primijenjena strategija prihrane supstratom omogućila je učinkovitije i ravnomjernije miješanje podloge za uzgoj. Kao posljedica postignute su velike koncentracije lipida u podlozi od 26,48 g L-1, udjel lipida od 269,25 mg g-1 i produktivnost od 2,04 g L-1 d-1. |

Parallel abstract (English) | Yeast Trichosporon oleaginousus was used for the simultaneous saccharification and lipid production process with pretreated corn cob with liquid to solid ratio (LSR) from 20:1 to 8:1 mLg-1. After 144 hours of batch cultivation, lipid productivity increased with the increase of LSR to the highest value of 1,69 gL-1d-1 at the LSR of 8:1 mLg-1. The highest lipid concentration of 11,5 gL-1 was obtained at LSR of 8:1 mL g-1 after 312 h, while the highest lipid content in total mass containing cells and unhydroyzed substrate of 172,8 mgg-1 was obtained at the LSR of 10:1 mL g-1 after 240 h. Furthermore SSF fed-batch additions of substrate 2,5 and 5 g. Batch substrate additions resulted in cumulative LSR equivalent to 5:1 and 6,7:1 mLg-1. The applied strategy of substrate addition enabled more vigorous and uniform stirring of cultivation media. This in turn resulted in high lipid concentration, lipid content and productivity of 26,48 gL-1, 269,25 mgg-1 and 2,04 gL-1 d-1, respectively. |
