Abstract

Kulture životinjskih stanica upotrebljavaju se u svim područjima biotehničkih znanosti. Rast, metabolizam i proliferacija stanica osigurana je dodatkom seruma koji pruža stanici sve potrebne nutrijente. Primjena seruma ima brojne nedostatke, a razvojem procesa s kulturama životinjskih stanica porasli su zahtjevi za smanjenjem upotrebe seruma te razvoja medija bez dodatka seruma. Cilj ovog rada bio je ispitati učinak različitih koncentracija seruma na rast stanične linije HeLa te utrošak glukoze u mediju za uzgoj. Ispitan je i učinak dodatka hidrolizata soje na rast HeLa stanica te mogućnost zamjene dijela seruma hidrolizatom soje. Medij s dodatkom 10% seruma pokazao se kao najbolji za rast i prinos HeLa stanica, a medij s 0,5% seruma pokazao je najsporiji rast i najniži prinos. Dodatak 5 g/L hidrolizata pri različitim volumnim udjelima FBS pokazao je da se ne može koristiti kao zamjena za proteine sadržane u serumu, izuzev medija s 10% FBS.

Abstract (english)

Animal cell cultures are used in many fields of biotechnical science. Growth, metabolism and proliferation of cells are ensured by using a serum which provides the cells with all necessary nutrients. Development of animal cell culture and many disadvantages of serum using resulted in demands to find a suitable medium without serum. The purpose of this work was to test the effects of different serum concentrations on the growth of HeLa cell line and the consumption of glucose in the medium. The effect of soy hydrolysate and its possibility to replace a part of a serum was also tested. It was shown that the best medium for cell growth and cell yield was obtained with 10% serum while media containing 0.5% of serum resulted in the lowest cell growth and yield.
