Abstract

Oksidacijski stres je poremećaj ravnoteže između stvaranja slobodnih radikala i reaktivnih metabolita i njihove eliminacije zaštitnim mehanizmima antioksidansa. Reaktivni kisikovi spojevi (ROS) mogu nastati egzogenim i endogenim putem. Važni endogeni izvori uključuju mitohondrije, metabolizam citokroma P450, peroksisome, mikrosome, aktivaciju stanica uključenih u upalni odgovor, sustav monooksigenaza, sintaze dušikova oksida i još neke enzime uključene u proces upale. Egzogeni izvori uključuju tvari iz okoliša i ksenobiotike. Oksidacijski stres može rezultirati biomolekulskim oštećenjem lipida, proteina i DNA. Poliklorirani bifenili (PCB) su aromatski sintetski organski spojevi kemijske formule C12H10-nCln koji pripadaju grupi spojeva poznatoj kao perzistentni organski zagađivači. PCB mogu inducirati nastajanje ROS koji su poznati ''okidači'' za oksidacijski stres. Takvo stanje dovodi do staničnih oštećenja, mutageneze, karcinogeneze i smrti stanice. U ovom radu preliminarno je ispitan mogući protektivan, antioksidacijski učinak vitamina E pri tretmanu CHO-K1 stanica sa dva odabrana, strukturno različita kongenera PCB-a – PCB 77 i PCB 153. Toksični učinak PCB 153 znatno je umanjen kada su stanice bile predinkubirane vitaminom E, za razliku od tretmana s PCB 77 gdje protektivni efekt vitamina E nije bio izražen.

Abstract (english)

Oxidative stress is an imbalance between production of free radicals and reactive metabolites and their elimination by protective mechanisms of antioxidants. Reactive oxygen species (ROS) can be produced from both endogenous and exogenous substances. Endogenous sources include mitochondria, cytochrome P450 metabolism, peroxisomes, microsomes, inflammatory cell activation, monoxygenase system, nitric oxide synthase and other enzymes involved in the inflammatory process. Exogenous sources include environmental agents and xenobiotics. Oxidative stress can induce biomolecular damage to lipids, proteins and DNA. Polychlorinated biphenyls (PCB) are aromatic synthetic organic compounds with the chemical formula C12H10-nCln and they belong to a group of compounds known as persistent organic pollutants. PCB can induce the generation of ROS which are known to ''trigger'' oxidative stress. Such a condition leads to cellular damage, mutagenesis, carcinogenesis and cell death. This preliminary thesis examines the possible protective, antioxidant effect of vitamin E on CHO-K1 cells exposed to two selected structurally different PCB congeners - PCB 77 and PCB 153. Pre-treatment of cells with vitamin E significantly reduces PCB 153 induced toxic effect, but shows no effect on PCB 77 exposed cells.
