Abstract

Peptidomimetici su spojevi čiji esencijalni farmakoforni elementi oponašaju 3D strukturu prirodnog peptida i/ili proteina te zadržavaju njegovu sposobnost interakcije s biološkim ciljanim sustavima, pri čemu zadržavaju jednak ili unaprijeđen biološki učinak. Budući da su peptidomimetici poznati kao potencijalni antitumorski agensi, u ovom radu je ispitana in vitro biološka aktivnost četiri različita ferocenska peptidomimetika na humanim tumorskim staničnim linijama (HeLa i MCF-7) te je određen mehanizam njihovog citotoksičnog djelovanja protočnom citometrijom. Sva četiri ispitana ferocenska peptidomimetika pokazala su inhibitorni učinak na rast staničnih linija, pri čemu peptidomimetik 1 ima najveći inhibitorni učinak na proliferaciju stanica. Također, određivanjem tipa stanične smrti u uzorcima tretiranim ispitivanim spojevima primjenom protočne citometrije je pokazano da peptidomimetik 1 ima najveći potencijal za induciranje apoptotične smrti stanica od svih ispitivanih peptidomimetika. Dobiveni rezultati ukazuju na mogući antitumorski učinak ispitivanih ferocenskih peptidomimetika.

Abstract (english)

Peptidomimetics are compounds whose essential pharmacophore elements mimic a natural peptide or protein in 3D space and which retain the ability to interact with the biological target and produce the same or even pronounced biological effect. They are known to be potential antitumor agents so in this research, in vitro biological activity of four different ferrocene peptidomimetics on human tumor cell lines (HeLa and MCF-7) and the mechanism of their cytotoxic activity using flow cytometry were determined. Based on the obtained results, their impact on people could be determined. All tested peptidomimetics showed an inhibitory effect on cell lines growth and peptidomimetic 1 showed the greatest inhibitory effect on cell lines proliferation. The results have also shown that the greatest inductor of apoptotic cell death is peptidomimetic 1. The obtained results indicate that these ferrocene peptidomimetics could have a possible antitumor effect.
