Abstract

Ljubičaste nesumporne bakterije su skupina bakterija koje imaju veliki potencijal u raznim područjima primjene, od industrijskog u kojem se najviše ističe proizvodnja biovodika do područja medicinske primjene za proizvodnju biokemikalija. U ovome radu je proveden uzgoj bakterije Rhodovulum adriaticum DSM 2781 u kojem je ispitivano na koji će način pojedini inhibitor i u kojoj koncentraciji utjecati na rast te bakterije. Kao inhibitori su korištene octena i mravlja kiselina, a rezultati su pokazali da do inhibicije dolazi bez obzira je li dodana octena ili mravlja kiselina te u kojoj je koncentraciji (1, 2, 3, 4 ili 5 g/L) dodana. Također, praćena je i sinteza pigmenata, s naglaskom na bakterioklorofil a koji se sintetizirao u bitno nižoj koncentraciji nego što je to bez dodatka inhibitora. Najveći prinos biomase zabilježen je kod koncentracije octene kiseline od 1 g/L te je iznosio 0.4279 g/L, a bakterioklorofil a se u najvećoj koncentraciji sintetizirao dodatkom 1 g/L mravlje kiseline te je njegova koncentracija iznosila 1.6589 mg/L.

Abstract (english)

Purple non-sulphur bacteria are a group of bacteria that have great potential in various fields of application, from industrial in which the most prominent one being production of biohydrogen to the field of medical application in production of biochemicals. In this thesis cultivation of the bacterium Rhodovulum adriaticum DSM 2781 was performed, in which it was investigated how a particular inhibitor and in what concentration will affect the growth of this bacterium. Acetic and formic acid were used as inhibitors and the results showed that inhibition occurs regardless of which acid (acetic or formic) was added and in what concentration (1, 2, 3, 4 or 5 g/L). Also, the synthesis of pigments was monitored, with emphasis on the bacteriochlorophyll a that was synthesized in a significantly lower concentration than in the absence of inhibitors. Biomass yield was shown to be highest at the mass concentration of acetic acid 1 g/L and was 0.4279 g/L while the highest concentration of bacteriochlorophyll a was synthesized by addition of 1 g/L of formic acid and its concentration was 1.6589 mg/L.
