Abstract

U ovom su radu istraženi fenolni profil i biološki učinci meda obične planike (Arbutus unedo L.) i njegove glavne bioaktivne komponente homogentizinske kiseline (HGA). Ukupni biološko aktivni spojevi (fenoli i flavonoidi) i antioksidacijski potencijal meda određeni su spektofotometrijski, a pojedinačni spregom tekućinske kromatografije ultravisoke djelotvornosti i spektrometrije masa visokog razlučivanja (UHPLC-HRMS). Ispitani su in vitro biološki učinci (antioksidacijski, geno-/citotoksični i geno-/citoprotektivni) meda i HGA na humanim staničnim modelima karcinoma pločastih stanica jezika (Cal27), hepatocelularnog karcinoma jetre (HepG2), adenokarcinoma epitela debelog crijeva (Caco-2) i limfocitima. Rezultati su ukazali na visok udio fenolnih spojeva i snažan antioksidacijski kapacitet meda obične planike. Detektirano je 56 fenolnih spojeva, što upućuje na složeni sastav meda. Primjenom mikronukeus testa i analizom kromosomskih aberacija utvrđena je visoka biokompatibilnost HGA, meda i njegova ekstrakta s limfocitima iz ljudske periferne krvi te njihov zaštitni učinak na razini citogenetičkih oštećenja izazvanih irinotekanom. Testirani spojevi pokazali su nisku citotoksičnost prema svim ispitivanim staničnim linijama. Stoga, med obične planike predstavlja značajan fitoterapeutik zbog visokog cito/genoprotektivnog i niskog citotoksičnog učinka.

Abstract (english)

This study investigated the phenolic profile and biological effects of strawberry tree honey (Arbutus unedo L.), as well as its main bioactive components homogenisic acid (HGA). Total content of bioactive compounds (phenols and flavonoids) and antioxidant potential of honey were determined using spectrophotometry, and individual phenolics using ultra-high performance liquid chromatography coupled to high-resolution mass spectrometry (UHPLC-HRMS). The in vitro biological effects (antioxidant, geno- / cytotoxic and geno- / cytoprotective) of honey and HGA were investigated on human cell models of tongue squamous cell carcinoma (Cal27), hepatocellular carcinoma of the liver (HepG2), epithelial colorectal adenocarcinoma cells (Caco-2) and lymphocytes. The results showed a high content of phenolics and a strong antioxidant capacity of strawberry tree honey. A total of 56 phenolic compounds were detected, indicating the honey’s complex composition. Using the micronucleus test and analysis of chromosomal aberrations, high biocompatibility of HGA, honey and its extract with lymphocytes from human peripheral blood and their protective effect on the level of cytogenetic damage caused by irinotecan were determined. The tested compounds showed low cytotoxicity to all of the cell lines tested. Therefore, strawberry tree honey is an important phytotherapeutic due to its high cyto-/ genoprotective as well as low cytotoxic effect.
