Učinak proteinskih hidrolizata konoplje i lana na rast, metabolizam i produktivnost CHO DP-12 stanične linije

Abstract

Fetalni životinjski serum najčešća je hranjiva dopuna mediju za uzgoj životinjskih stanica. Zbog njegovog porijekla te drugih ograničenja seruma, već više od trideset godina ispituju se zamjenski supstrati poput hidrolizata proteina biljaka. Cilj ovog rada je istražiti in vitro utjecaj proteinskih hidrolizata uljne pogače lana i konoplje na rast, vijabilnost, metabolizam i produktivnost CHO DP-12 stanične linije uzgajane u suspenziji. Pritom je ispitan utjecaj peptidnih frakcija hidrolizata veličine do 10 i do 1 kDa u koncentracijama od 0,5 i 2 g Lˉ¹. Rezultati su pokazali da su ispitane frakcije hidrolizata proteina konoplje imale pozitivan učinak na produktivnost, a da na rast puno bolje utječe koncentracija hidrolizata od 0,5 g Lˉ¹, dok koncentracija od 2 g Lˉ¹ djeluje citotoksično na stanice. Frakcije proteina <1 kDa hidrolizata lana vrlo slabo utječu na rast i proliferaciju, no pozitivno utječu na produktivnost stanica, dok frakcije proteina <10 kDa hidrolizata lana pozitivno utječu na rast i proliferaciju stanica, ali negativno utječu na njihovu produktivnost.

Abstract (english)

The usual supplement to animal cell culture medium is fetal animal serum. Due to serum's compositional shortcomings and safety concerns, the use of plant protein hydrolysates has been investigated as a potential alternative to the serum for more than thirty years. The aim of this paper is to investigate the in vitro effect of protein hydrolysates of flaxseed and hemp on growth, viability, metabolism and productivity of CHO DP-12 cell line grown in suspension. The influence of peptide fractions of hydrolysates up to 10 and up to 1 kDa in concentrations of 0.5 and 2 g Lˉ¹ was investigated. The results showed that the tested fractions of hemp protein hydrolysates had a positive effect on productivity and that growth was much better influenced by a hydrolysate concentration of 0.5 g Lˉ¹, while a concentration of 2 g Lˉ¹ has a cytotoxic effect on cells. Protein fractions <1 kDa of flax hydrolysate have very little effect on growth and proliferation, but have a positive effect on cell productivity, while protein fractions <10 kDa of flax hydrolysate have a positive effect on cell growth and proliferation, but negatively affect their productivity.
