Proizvodnja celulolitičkih enzima s plijesni Trichoderma reesei i optimizacija enzimske hidrolize kukuruznog oklaska

Abstract

Plijesan Trichoderma reesei korištena je za proizvodnju celulolitičkih enzima na polučvrstoj i čvrstoj podlozi s kukuruznim oklaskom kao izvorom ugljika. Kako bi se postigla maksimalna celulolitička aktivnost (FPA) izvršena je optimizacija hranjivih podloga. Primjenom filter papir testa, maksimalna FPA vrijednost od 6,2845 IU/g glukana dobivena je u 28. danu uzgoja plijesni na čvrstoj podlozi s neobraĎenim kukuruznim oklaskom, pšeničnim mekinjama i 70 % vode. Na polučvrstoj podlozi, maksimalna FPA vrijednost od 5,3710 IU/g glukana, postignuta je u 9. danu uzgoja. Polučvrsta hranjiva podloga sadrţavala je obraĎen kukuruzni oklasak, pšenične mekinje, otopinu Tween 80 te liofilizirane celulaze iz T. reesei. Nadalje, istraţen je učinak dodatka Tweena 80 na kinetiku enzimske hidrolize i hidrolizu strukturnih ugljikohidrata na podlogama s dvjema različitim koncentracijama supstrata, 10 g L-1 glukana i 100 g L-1 kukuruznog oklaska. Ispitano je sedam različitih koncentracija Tweena 80 (0, 1, 2, 4, 6, 12 i 20 g L-1). Najveće koncentracije glukoze i ksiloze postignute su dodatkom 12 g L-1 Tweena 80. Rezultati su pokazali da Tween 80 povećava brzinu enzimske hidrolize i učinkovitost procesa enzimske hidrolize strukturnih ugljikohidrata.

Abstract (english)

Fungus Trichoderma reesei was used for the production of cellulolytic enzymes in the semi-solid and solid media with corncobs as carbon source. In order to achieve maximal cellulolytic activity (FPA), the nutrient composition of media was optimised. Using filter paper assay, maximal FPA of 6.2845 IU/g glucan was determined at the 28th day of the cultivation in the solid medium with raw corncobs, wheat bran and 70 % water. In semi-solid medium the maximal FPA of 5.3710 IU/g glucan media, was achieved after 9 days of cultivation. The semi-solid medium contained the pretreated corncobs, wheat bran, Tween 80 and lyophilized cellulases from T. reesei. Furthermore, the effect of Tween 80 concentration on kinetics of enzymatic hydrolysis and hydrolysis of structural carbohydrates was determined in media containing two different concentrations of substrate, 10 g L-1 glucan and 100 g L-1 corncobs. Seven Tween 80 concentrations (0, 1, 2, 4, 6, 12 and 20 g L-1) were examined. The highest concentration of glucose and xylose was obtained at 12 g L-1 Tween 80. The results showed that Tween 80 increased the hydrolysis rate and efficiency of enzymatic hydrolysis of structural carbohydrates.
